Objective To determine the prevalence of coagulation factor XIII Val34Leu gene polymorphism and FXIII plasma level in Han people in Shanghai and to investigate whether it is associated with the risk of pulmonary thromboembolism (PTE).Methods Fifty PTE patients and seventy age and sex-matched healthy controls from the same geographic area were enrolled in the study. All patients were diagnosed on the basis of lung ventilation/perfusion scan and/or multi-slice CT pulmonary angiography as well as medical history and clinical manifestations.Polymerase chain reaction (PCR), restriction enzyme digestion and Polyacrylamide Gel Electrophoresis (PAGE) were used to identify the FXIII Val34Leu gene polymorphism.The plasma level of FXIII antigen was measured by sandwich enzyme linked immunosorbent assay.Results FXIII Val34Leu gene polymorphism was not found in both PTE patients and controls. No difference was found in plasma FXIII antigen level between the PTE patients and controls.Conclusion There is no definitely correlation between FXIII Val34Leu gene polymorphism,plasma FXIII antigen level and PTE risk in Han people in Shanghai.
