Objective:To determine the effect of mineral trioxide aggregate (MTA),glass ionomer cement (GIC) on the proliferation and differentiation of MG-63 cells.Methods:Prepare the leaching liquor of MTA and GIC of three different concentrations (40mg/ml,20mg/ml,10mg/ml). The 3rd generation logarithmic phase of the MG-63 cells were seeded in 96-well plates and 24-well plates, by adding different leaching liquor and then cultivant, using DMEM culture medium as control group, the proliferation and differentia- tion of MG-63 cells and alkaline phosphatase (ALP) activity was measured after 72 hours of incubation. SPSS12.0 software package was used to make statistical analysis.Results:The optical density (OD) readings of each concentration of MTA leaching liquor were all higher than control group.And the values were associated with the concentration series. The higher concentration was, the higher OD values were. Compared each concentration of group and control group with one-way ANOVA, there was a significant difference. Compared the ALP activity,the OD values of each concentration of MTA group were all higher than the control group. And the OD value raised with the MTA concentration. statistically significant differences were found between the control group and 20mg/ml and 40mg/ml concentration group; GIC Group of the concentration of OD values lower than those in the control group, with the increase in the concentration of GIC extraction, OD values decreased, the differences between 40mg/ml concentration group and control group were statistically significant. Conclusion:1. MTA stimulates the proliferation and of MG-63 cells, It is a material with good biocompatibility; but GIC fail to stimulates the proliferation of MG-63 cells.2. MTA can increase alkaline phosphatase activity, that is the function of promoting cell differentiation; but GIC fail to increase alkaline phos- phatase activity.
