Objective:To investigate the expression levels of platelet-derived growth factor-BB(PDGF-BB) and its role in the pathogenesis of the patients with hepatic diseases.Methods:1.The expression of PDGF-BB in 24 cases of primary HCC tissues and 18 cases of normal liver tissues were detected by immunohistochemistry.2.The expression levels of serum PDGF-BB in 30 patients with chronic hepatitis B virus,n=26;hepatitis C virus,n=4),24 patients with posthepatitic cirrhosis,30 patients with primary carcinoma of liver and 20 normal controls(NCs) were measured by enzyme linked immunosorbent assay(ELISA).The levels of PDGF-B mRNA in peripheral blood mononuclear cells were measured by reverse transcription polymerase chain reaction(RT-PCR).Results:1.The positive expressions of PDGF-B was 66.7%in HCC,5.6%in normal liver tissues,respectively.There was significant differences between the two groups.2.The serum levels of PDGF-BB and the levels of PDGF-B m RNA in peripheral blood mononuclear cells were significantly higher in the patients with liver injury[chronic hepatitis,posthepatitic cirr hosis,Primary carcinoma of liver was(108.35±19.96),(240.38±16.84),(376.57±22.52),(28.36±8.35) pg/ml],and(0.3753±0.0 29 2),(0.6029±0.0333),(0.7563±0.0438) respectively]than tho se in NCs[(28.36±8.35) pg/mL,(0.2275±0.0384),F=1774.40,1 037.42,P<0.01]there was close correlation between PDGF-BB lev el and hepatic function level in liver injury.Conclusions:1.PDGF-B exhibited low expression in normal liver tissue,but high in HCC.2.In the patients with hepatic diseases,the levels of PDGF-BB,PDGF-B mRNA were significantly increased as compared with that of NCs,while the levels of PDGF-B in HCC was higher significantly than that in posthepatitic cirrhosis,the levels of PDGF-B in posthepatitic cirrhosis significantly is higher than that in chronic hepatitis.PDGF-BB may play an important role in promoting inflammatory reactions leading to massive liver damage in the patients with hepatic diseases.The levels of PDGF-BB and PDGF-B mRNA can be used to evaluate the degree of liver damage,and for early diagnosis and prognosis.
