Experimental Study On The Effect Of Tian Ma Granules On Proliferation Of Human Intestinal Cancer Cell Line SW-480 Transplantated Into Subrenal Capsul Of Mouse

Objective: To study the effect of Tian Ma Granules on proliferation of Human intestinal cancer cell and preliminarily explore its mechanism.Methods: (1) model rats of transplantation tumor of human intestinal cancer cell line SW-480 were established. The Tian Ma Granules, the blank control group, 5-Fu positive control group and the combined medication group were set up. (2)After 8 days medication of Tian Ma granules through gastrogavage, the antitumor effect of each group was compared. (3) The transplantation tumor karyon morphology and the gene expression of Ki-67,PCNA in each group were quantitative detected and compared by the application of pathomorphology, immunohistochemistry and image analysis technique.Results: (1) The Tian Ma Granules could inhibit the growth of transplantation tumor. The tumor 's weight or volum in Tian Ma Granules group was lower than that in the blank group(P < 0.01). The tumor's weight or volum in the combined medication group was obviously lower than that in the blank group and that in the 5-Fu positive control group(P < 0.01).(2) Tian Ma Granules could improve the cell and karyon morphology of transplantation tumor, and reduce the physical content in nucleus(P < 0.01). This effect was most significant in the combined medication group. The difference was significant compared with the blank group and 5-Fu positive control group(P < 0.01). (3) The Ki-67 count in Tian Ma Granules group was lower than that in the blank control group(P < 0.01), and that in the combined medication group was lowest, furthermore the difference was significant compared with the 5-Fu positive control group(P < 0.05). Tian Ma Granules could depress the level of PCNA expression, and this difference between the combined medication group and 5-Fu positive control group was significant(P < 0.05).Conclusion: (1) Tian Ma Granules has definite antitumor effect against human colonic carcinoma, and this effect is similar to that of the 5-Fu.(2) Tian Ma Granules can inhibite the cell proliferation of human intestinal cancer, which may be correllated with its effect of improving karyon morphology and reducing the physical anabolism in nucleus and depressing the expression level of PCNA and Ki-67.(3)The combined application of Tian Ma Granules and 5-Fu has synergistic effect.
