| Objective:The chief contribution of Ubiquitin Proteasome Pathway(UPP) is to degradate paraprotein,and sustain the physiological function of cells.At present, mang hereditary disease and acquired disease are concerned with the functional disorder of UPP.We investigate the expression of peripheral blood lymphocyte ubiquitin mRNA in chronic hepatitis B,active cirrhosis,severe hepatitis,and evaluate its clinical significance.Methods:Ninety six patients were enrolled in the study,whom were divided into 3 groups:chronic hepatitis B group(40 cases),male 34 cases,female 6 cases, average age 35.85 years old(19~70 years old);active cirrhosis group(30 cases), male 25 cases,female 5 cases,average age 51.52 years old(31~68 years old);and severe hepatitis group(26 cases),male 23 cases,female 3 cases,average age 40.69 years old(26~59 years old);The patients who were inpatients in Jinan infectious disease hospital from August 2007 to January 2008,and in line with the diagnosis and typing standard in 2000 on Tenth National Symposium on Viral Hepatitis.In addition,the healthy control group(26 cases) was established,who were from Richard Body Center of Jinan infectious disease hospital,male 22 cases,female 4 cases,average age 40.42 years old(19~70 years old);The ubiquitin mRNA of peripheral blood lymphocyte was detected by real time PCR,as well as intra-referenceβ—actin.Differences among groups were analysed by one-way ANOVA.Electrochemiluminescence was used to quantitate serum HBeAg and anti-HBe;Fluorescent quantitation was applied to detect serum HBV-DNA.Relationships between ubiquitin mRNA and the indexes of serum virology were treated by correlation analysis.Results:1,The expression levels of peripheral blood lymphocyte ubiquitin mRNA in all studied groups are lower than the healthy control group(P = 0.00).2,The decreasing expression of ubiquitin mRNA was detected in chronic hepatitis B group,active cirrhosis group and severe hepatitis group(78.23±14.14%,70.34±15.78%,61.88±23.43%,respectively ).There is significant difference between chronic hepatitis B group and severe hepatitis group(P<0.05).3,the expression of peripheral blood lymphocyte ubiquitin mRNA is significantly negative correlation with the levels of HBeAg and HBV DNA(P<0.05),but no correlation with anti-HBe.Conclusions:1,The ubiquitin mRNA of peripheral blood lymphocyte was detected by real time PCR and 2^-△△CT,which were simple and reliable.2,This study suggests that the expression of peripheral blood lymphocyte ubiquitin mRNA is insufficient in all patients,the lower expressive the severer condition.3,There is relationship between ubiquitin mRNA expression and the copies of hepatitis B,the higher copies the lower expression.4,The expression abnormity may be involved to occurrence and development of the disease. |
