Objective:To investigate the expression of TIP30 and CD34-labeled microvessel density(MVD) in gastric cancer tissues and para-cancer tissues,to analyze the role of TIP30 in the genesis and development of gastric cancer and its impact with angiogenesis.Methods:52 gastric cancer tissues and 47 para-cancer tissues which have been cut via operation were choosed from our hospital.The expression of TIP30 and CD-34 labeled MVD were measured by immuohistochemical method,and the level of TIP30mRNA was measured by RT-PCR both in gastric cancer tissues and para-cancer tissues。And next analyze the difference of expression of TIP30 and CD-34 labeled MVD in gastric cancer tissues and para-cancer tissues。And analyze relationships between the two index between age,sex,cell differentiation and size of carcinoma,lymph node metastasis and TNM stage。Statistical methods:All stastics were performed using SPSS 13.0 software,complete with statistics significance(p<0.05)Results:1.Bytheresults of immunohistochemistry,the TIP30 protein was Mainly expressing in the cytoplasms and cell membranes, brownish-yellow,graininess or mass.There were 28 positive expressions of TIP30 protein in all 52 gastric cancer tissues and 40 positive expressions in 47 para-cancer tissues.The positive rate were53.8%and 85.1%respectively.The expressions of TIP30 protein in gastric cancer tissues were much lower than in para-cancer tissues,there was obvious difference(x~2=11.2,P=0.001).By the results of RT-PCR,43 expressions ofTIP30mRNA were detected in all 52 gastric cancer tissues(82.7%),and 38 expressions were detected in all 47 para-cancer tissues(80.9%).Refer to theβ-actin,the level of TIP30mRNA was 0.67±0.24 in gastric cancer tissues,and 0.72±0.20 in para-cancer tissues,there were no obvious difference in the positive rate and level of TIP30mRNA between the two kinds of tissues(x~2=11.2,P=0.813/ t=0.732,P>0.05).2.The expression of CD34 in tissues.The MVD in cancer tissues and para-cancer tissues were 27.37±2.68 and 21.87±4.11 respectively,and the MVD in cance tissues was obviously higher. (t=2.862,p<0.01)3.By the results of immunohistochemistry,the expression of TIP30 protein in gastric cancer was observed to correlate with lymph node metastasis and TNM stage,expression of TIP30 protein was not related with age,sex,cell differentiation and size of carcinoma.By the results of RT-PCR,the levels of TIP30mRNA were not related with age,sex and cell differentiation,but levels of TIP30mRNA were relatively lower in the bigger,later TNM stage tissues and tissues with lymph node metastasis.(p>0.05)4.The CD34-labeled MVD was not related with age,sex,cell differentiation,and was Related with size of carcinoma,lymph node metastasis and TNM stage,and was related With expression of TIP30.The MVD was 28.54±2.81 in 24 TIP30-negative cancer tissues and 26.36±2.13 in the 28 TIP30-positive cancer tissues.There was obvious difference(t=-3.18,p=O.O03 ).5.The analysis of relevance between the enumeration of MVD and the expression of TIP30.Referring to the staining results of immunohistochemistry,the expression levels of TIP30 were figured as such points:0 point,1point,2point,3point,4point,6point and 9 point,The analysis of relevance was carried out by Speraman rank correlation,it shows that the points of TIP30 have negative correlation with MVD(r=-0.777,p<0.01).Conclions:1.The expression of TIP30 protein in gastric cancers were much lower than in para-cancer tissues,while there was no significant difference in the level of TIP30mRNA between the two kinds of tissues.And its expression was related with lymph node metastasis and TNM stage.This reveals that TIP30 participates in the genesis and Development of gastric cancer,but it may mainly function in the course of post-transcription and translation2.The MVD can reflect the vessel formation of tumor tissues. The MVD in TIP30-positive tissues was much lower than in negative tissues.And the points of TIP30 have negative correlation with MVD.This reveals that the effect of TIP30 gene for tumor genesis and growth may be partially completed through depression of vessel formation.3.The expression of TIP30 and the enumerations of MVD had significant relations with the tumors' clinical features,such as genesis,invasion and lymph node metastasis.So they can be used as an important index for the capacity of growth,invasion and metastasis in the gastric carcinomas,and be propitious to tumor diagnosis,treatment and prognosis.
