Objective To analyze the association among Eales disease and histocompati-bility leukocyte antigen(HLA-A/B,HLA-DRB/DQB) alleles and tuberculous infection, and to explore susceptible genes and protective genes associated with Eales disease and tuberculous infection in a population Han nationality from ZUN YI city in Guizhou province. Methods The subjects were analyzed by case-control study consisted of three groups such as Eales disease group(47 cases),pulmonary tuberculosis group(36 cases) and normal control group(100 cases).Eales disease group was divided four part.Part one, 12 cases who had suffered from pulmonary tuberculosis.Part two,27 cases who had not suffered from pulmonary tuberculosis.Part three and four were purified tuberculin test positive and negative, the cases were 27 and 12 respectively.DNA samples from 47 patients with Eales disease,36 patients with pulmonary tuberculosis and 100 healthy people were detected by polymeriase chain reaction with sequence-specific primers(PCR-SSP).The alleles of HLA-A/B and HLA-DRB/DQB from Eales disease group were compared with those from tubercle group and normal control group, and the correlativity test of common susceptible genes were made to analyzed the potential relationship between Eales disease group and pulmonary tuberculosis group. Results The frequency distribution of HLA-A~*02(P=0.000,OR=9.719, OR95%CI:4.377-21.580),HLA-B~*07(P=-0.001,OR=11.605;OR95%CI:2.397-56.191) alleles in Eales disease was obviously higher than control, but the allales of HLA-A~*11 (P=0.048,OR=0.495,OR95%CI=0.245-1.000) were obviously lower than control(P<0.05),two groups had significant diference(P<0.05).The frequency distribution of the allales in part one and two demonstrated that HLA-A~*02(X~2=0.290,OR=1.455,OR95%CI =0.371-5.709,P=0.590),HLA-A~*11(X~2=0.395,OR=1.545,OR95%CI=0.398-6.035,P=0.530) and HLA-B~*07(X~2=0.157,OR=0.700,OR95%CI=0.119-4.104,P=1.000) alleles had not significant diference(P>0.05) in two groups. The frequency distribution of the allales in part three and four demonstrated HLA-A~*02(X~2=0.29,OR=0.688,OR95%CI=0.175-2.699, P=0.59),HLA-A~*11(X~2=0.001,OR=1.027,OR95%CI=0.257-4.108,P=0.970) and HLAB~*07 (X~2=0.157,OR=0.700,OR95%CI=0.119-4.104,P=1.000) alleles had not significant diference(P>0.05) in two groups. The frequency distribute of HLA-A/B-,DRB/DQB in the Eales disease group, pulmonary tuberculosis group and control group demonstrated that HLA-A~*A02(X~2=58.225,P=0.000),HLA-A~*24(X~2=7.762,P=0.025),HLA-A~*68 (X~2=8.257,P=0.037),HLA-B~*07(X~2=16.390,P=0.001) andHLA-DRB~*16(X~2=7.765, P=0.047) alleles had significant diference(P<0.05) in three groups. The frequency distribution of HLA-A~*24(OR=4.238,OR95%CI=1.416-11.521,P=0.007) in pulmonary Tub -erculosis group was lower than Eales disease group, two groups had significant diference (P<0.05),but in HLA-A~*02 alleles had not significant diference(P>0.05) in two groups. Conclusion The results showed that the alleles of HLA ~*02,-B~*07 may associate with an antagonist effect in Eales disease, but HLA-A~*11 may be a protective gene of this disease, The alleles of HLA-A~*02 may be common susceptible genes of Eales disease and pulmonary tuberclosis.HLA-A~*11and HLA-A24 were protective gene of Eales disease and pulmonary tuberculosis respectively. In a word, the frequency distribution of susceptible genes with Eales and pulmonary tuberculosis in a population Han nationality from ZUN YI city in Guizhou province had not significant diference.
