SPA And Sperm DNA Integrity Detection For IVF-ET

ObjectiveWith the development of assisted reproductive technology,the asseament of sperm function has come to cellular and molecular level from traditional semen analysis. The zona-free hamster oocyte sperm penetration assay permits the observation of various events related to fertilization as the function of capacitation,acrosome reaction,sperm-egg fusion,male pronucleus forming.The intergrity of sperm DNA is well associated with the fertility capability,cleavage of fertilized ovum and develepment of embryo.The aim is to dicuss the value of zona-free hamster oocyte sperm penetration assay and DNA fragmentation detection for IVF-ET.MethodsA total of 41 men in infertile couples opting for IVF-ET from reprodutive medical research center,the first hospital of Lanzhou university,were derided into two groups of completely male sterility and female infertility depending on their factors of infertility.They were assessed by analysing male ages,fertility index,rates of penetration,DNA fragmentation,IVF fertility,clearvage,good quality embroyo, embroyo nidation and clinic pregnant,and the correlation between the rates of SPA penetration and sperm DNA fragmentation with IVF fertility,clearvage,good quality embroyo,embroyo nidation respectively.ResultsTwo groups have significant differences in penetration rates,fertility index,DNA fragment rates(P＜0.05),while no differences in male ages,the rates of IVF fertility, clearvage,good quality embroyo,nidation and clinic pregnant(P＞0.05).In grounps of male sterility and female infertility,conspicuous correlation was observed in penetration rates with the rates of IVF fertility,clearvage and good quality embroyo,DNA fragment rates with the rates of IVF fertility,clearvage and good quality embroyo respectively(P＜0.05).ConclutionSperm DNA integrity detection has a good privilege with routine semen analysis in forecasting the outcome of ART,assessing male infetility,selecting the optimal way for ART and assisting to choose suitable scheme for optimization,freezing and minimizing iatrogenic damage.Therefore,in clinical,only combining zona-free hamster oocyte sperm penetration assay,sperm DNA integrity detection with other functional and characteristic inspection systems can make an allround evaluation for male fertility.
