[objective]In order to find out the condition of male semen in Yunnan and to explore the factors affecting male semen quality in Yunnan province,7046 male semen samples were analyzed.[Method]7046 male semen samples were collected from people who came to our hospital for fertility assessment in Yunnan province.They were divided into five groups according to age:Group A?25 years old,25 years old<Group B?30 years old,30 years old<Group C?35 years old,35 years old<Group D?40 years old,Group E>40 years old,according to the fifth edition "The WHO Laboratory Manual for the Examination and processing of human semen" as the standard,7046 male semen samples were analyzed.[Result]A total of 7046 semen samples,azoospermia accounted for 5.52%(389/7046).Among 6657 semen samples without azoospermia,the normal group accounted for 54.04%(3808/7046),the abnormal group accounted for 40.43%(2849/7046),the abnormal group accounted for 30.81%(2171/7046),asthenozoospermia accounted for 5.64%(398/7046),abnormal liquefaction time accounted for 0.37%(26/7046),oligospermia accounted for 0.2%(14/7046).With age increased,the proportion of asthenozoospermia showed an upward trend.The PR+NP,PR and semen volume of group E(>40 years old)were significantly lower than those of group A,group B,group C and group D(P<0.01),while sperm concentration and liquefaction time were significantly higher than group A,Group B,Group C,Group D(P<0.01).[Conclusion]The semen quality of males in Yunnan showed a downward trend with age.30-35 years old were the excessive stage of the decline of male semen quality in Yunnan.After 40 years old,the semen quality decreased significantly(P<0.01).Asthenozoospermia were the main manifestation of male semen abnormalities in Yunnan,and the proportion of asthenozoospermia increased with age.[Objective]By analyzing the correlation between the human semen humanin concentration sperm quality in order to explore the molecular mechanism of humanin affecting male semen quality.[Method]185 semen samples were collected,they were divided into four groups according to the CASA,including normospermia groups(n=59),oligospermia groups(n=32),asthenospermia groups(n = 68),and oligoasthenospermia groups(n = 26)Immunofluorescence were used to detected the expression of humanin in testis and spermatozoa.To investigate the correlation between humanin level in sperm and sperm parameters,ELISA was used to measured humanin level in spermatozoa.Methods as follow:?Ten samples were randomly selected form normospermia groups,oligospermia groups,asthenospermia group,sperm humanin were measured by ELISA.?All sperm samples in normospermia groups and asthenospermia group were measured by ELISA,then to analyze the correlation between humanin levels in spermatozoa and progressive sperm;?To compare the humanin level in progressive spermatozoa with humanin level in non-progressive/immotile spermatoza,12 samples were selected by the swim-up method.Enzyme-linked immunosorbent assay kit(ELISA)were used to detect the determination of seminal plasma humanin.Spearman method were used to analyze the correlation between seminal plasma and sperm parameters including sperm concentration,progressive sperm motility,VAP,VSL,VCL,ALH,BCF,LIN,STR,WOB.[Result]Hiimanin was abundantly expressed in human testis tissue,mainly expressed in Leydig cells and spermatocytes.In sperm,it was mainly expressed in the neck and middle part of the sperm,and the entire flagella of the sperm is also stained.Sperm humanin level in normospermia groupswere significantly higier than asthenospermia groups(P=0.01).And there was no significant difference between sperm humanin levels in oligospermia groups and normospermia groups(P=0.21).There was no significant difference between sperm humanin levels in oligospermia groups and asthenospermia groups(P=0.16).There was a significant correlation between sperm humanin levels in oligospermia groups,normospermia groups and progressive sperm motility(r=0.70,P<0.001).Humanin level in swim-up spermatozoa were significantly higher than non-swim-up spermatozoa(P=0.03)All seminal plasma humanin levels were between 10.87-285.lpg/ml,median was90.47pg/ml,25 th to 75 th percentiles were 58.5-128.6pg/ml.?humanin level in normospermia groups are significantly higher than oligospermia groups(P<0.001)? humanin level in normospermia groups are significantly higher than oligoasthenospermia groups(P<0.001)?humanin level in normospermia groups are significantly higher than asthenospermia groups(P<0.001).There was a significant correlation between hvimanin level in seminal plasma and sperm concentration(r=0.49,P<0.001).There was a significant correlation between humanin level in seminal plasma and progressive sperm motility(r=-0.29,P<0.009),there was a negatively significant correlation between humanin level in seminal plasma and STR(r=-0.20,P=0.009)and there was positively significant between humanin level in seminal plasma and ALH(r=0.17,P=0.03).There was no significant correlation between hiunanin level in seminal plasma and other parameters.[Conclusion]The study found that humanin was expressed in human testes and spermatozoa.Further research found that both SP humanin concentration and the humanin expression level in spematozoa were significntly positively correlated with sperm concentration and sperm motility,indicating fliat Humanin may participate in spermatogenesis regulation,especially for sperm motility.
