OBJECTIVE:For male patients with infertility couples undergoing in vitro fertilization-embryo transfer(IVF-ET)in our hospital,we explored the relationship between sperm DNA fragmentation index(DFI)and semen routine and sperm DNA fragmentation index and in vitro fertilization embryos.Correlation between clinical outcome events of transplantation(IVF-ET)technology.This is to evaluate the significance and value of DFI in assessing male fertility.METHODS:We collected semen specimens from male patients in our couple who had come to our hospital for the treatment of infertility in IVF-ET IVF between January 2017 and November 2018.These specimens were routinely examined for semen and sperm DNA fragmentation index(DFI)inspection.We tracked and recorded the entire in vitro fertilization process,monitored the development of embryos after fertilization,and used SPSS 19.0 data processing software to correlate DFI,routine semen test results,and statistics of in vitro fertilization and post-fertilization embryo development.Sexual analysis,and one-way analysis of variance for these sets of data.RESULTS:A total of 124 male patients with infertile couples were included in the study through our inclusion criteria.The conclusions of the experiment are as follows:1.Correlation analysis shows that sperm motility,forward motor sperm rate(PR),and non-forward motor sperm rate(NP)are negatively correlated with sperm DNA fragmentation index(DFI).Sexual relationship,that is,the higher the DFI value,the lower the sperm activity rate,PR,NP;the immobile sperm rate(IM)has a positive correlation with DFI,that is,the higher the DFI value,the greater the proportion of immobile sperm.2.According to the size of DFI,the three groups of ABC,group A(DFI?10%),group B(10%<DFI<30%),group C(DFI?30%),the regular parameters of each group of semen and their average ANOVA.Because the number of cases in each group is different,the welch test is required before the analysis of variance.The results of analysis of variance showed that sperm activity rate,PR,NP,and IM were significantly different under different DFI groups.However,unlike the above results,there was no significant difference in sperm concentration under the DFI grouping.In terms of immobility rate,the experimental results showed that the proportion of IM in group C was higher than that in group B,and the proportion in group A was the smallest,that is,group C>group B>group A.In terms of active sperm,the sperm activity rate,PR and NP in group A were significantly higher than those in group B,and group B was higher than group C,ie group C<group B<group A.This indicates that the sperm movement is most active when the sperm DNA fragments are small,and the sperm DNA movement rate is affected when the sperm DNA fragment rate is high,showing a significant increase in the number of immobile sperm.3.When calculating the fertilization rate,cleavage rate and excellent embryo rate under different DFI groupings,it was found that the data obtained in the three groups were statistically significant,and the P values were all less than 0.05.The three clinical outcone measures of fertilization rate,cleavage rate and excellent embryo rate showed significant statistical differences under different DFI groups.CONCLUSION:In addition to conventional semen analysis,sperm DNA fragmentation can be considered as another necessary option.Sperm DFI detection can be used as a reference option for IVF-ET technology fertilization outcomes.Reference to the level of sperm DFI during the diagnosis and treatment process can help improve the success rate of human assisted reproductive technology.
