ObjectiveDetect the expression of KAI1/CD82protein and MMP9protein in breastcancer tissue and benign breast tissue to explore its relationship withclinicopathological features,and analyze the correlation between the two,to lookfor the inhibition of tumor metastasis protein generesearch and individualtherapy to provide new information and to provide the new ideas of improvingthe treatment of malignant tumors and preventing further tumor.MethodsUsing immunohistochemical SP method to detect the expression ofKAI1/CD82protein and MMP9protein,120breast cancer cases and25breastbenign tissue cases in the First Affiliated Hospital of Liaoning Medical collegefrom January2000to January2001,which Breast Surgery surgical resectionand the pathologist to confirm the diagnosisment,and to analyze their correlationwith clinicopathological features,and to explore the relationship between the twoprotein genes;application by western blot to analyze the expression of theKAI1/CD82protein and MMP9protein,in April2011to in May2011,the collectionof12fresh breast cancer cases,benign breast tissue of4cases,and to explorethe expression of these two genes in the different stages(I,II and PhaseIII)ofbreast cancer and benign breast tissue;follow-up120cases of breast cancerpatients to analyze survival.Statistical data processing applications SPSS17.0software using the X2test for pairwise comparisons,ranked data using themethod of Spearman correlation analysis,survival curves using the KM method for analysis,inspection level of P <0.05was statistically significant.Results1、The protein of KAI1/CD82is mainly expressed in the cytoplasm and(or)cellmembrane and MMP9in the cytoplasm.the positive expression rate ofKAI1/CD82in120cases of breast cancer organizations was45.00%,thepositive expression rate is72.00%in25cases of benign breast tissue,thedifference was statistically significant(P<0.05).2、The expression of KAI1/CD82protein in lymph node metastasis,pathologictype,tumor size,clinical stage and histological grade are closely correct.Thelower histological grade,the higher clinical stage and pathologic stage,and thelower KAI1/CD82the positive expression rate,the difference was statisticallysignificant(P<0.05);while patients with menopausal status,age,HER-2,PR andER are not related(P>0.05).3、 The expression of MMP9protein with histological grade,lymph nodemetastasis,clinical stage,pathological type and PR are closely related tohistological grade,the lower histological grade,the higher pathologic stage andclinical stage,and the higher the expression of MMP9,the difference arestatistically significant(P<0.05);while patients with patient age,tumorsize,menopausal conditions and HER-2,ER,etc are not related(P>0.05).4、The positive expression of Both KAI1/CD82protein and MMP9protein is42cases,and6cases both negative for expression.KAI1/CD82protein-positive andMMP9protein negative expression is12cases.KAI1/CD82protein negative andpositive MMP9protein expression is60cases,the analysis of using statisticalSpearman method shows both a negative correlation(P=0.04,r=-0.18).5、KAI1/CD82high-expressing group of breast cancer after10-year survival ratewas100%,low expression of1,3,5,10-year survival rates were100%,79.17%,67.50%,54.17%,the difference was statisticallysignificant(P<0.05)between the two groups;MMP9low-expressing group of breast cancer after10-year survival rate was100%,high expression of1,3,5,10-year survival rates were100%,89.17%,76.67%,66.67%,the differencebetween the two groups was statistically significant(P<0.05).6、western blotting analysis the results of12cases of breast cancer tissues andfour cases of benign breast tissue suggest that:high expression of KAI1/CD82protein in benign breast tissue,and low expression of MMP9protein in benignbreast tissue;KAI1/CD82with breast cancer(I,II,PhaseIII)stages increased,andits expression gradually reduced,MMP9with the increased staging of breastcancer,and its expression was gradually increased.Conclusion1、The expression of KAI1/CD82protein and MMP9protein in breast cancermay be closely related to the occurrence and development of breast cancer.2、 The expression of KAI1/CD82protein in breast cancer suggests thatKAI1/CD82protein may be one of the mechanisms that suppress breast cancermetastasis.3、Found by the follow-up,high expression of KAI1/CD82protein and lowexpression of MMP9protein are good for the survival of the prognosis ofpatients.4、The expression of KAI1/CD82protein and MMP9protein of breast cancer isnegative correlation.5、The expression of KAI1/CD82and MMP9protein are associated with breastcancer invasion,metastasis and prognosis,with a certain reference value for theclinical judgment of the patient’s condition and treatment.
