ObjectiveCancer-related death is the second leading cause of death,after cardiovascular disease,and breast cancer is the second leading cause of death among all types of cancer;It is one of the leading causes of death in the Chinese population but also a significant cause of the high economic burden of Chinese families.Lamin B2 is one of the most representative subtypes of laminin type B.Encoded by the LMNB2 gene,Lamin B2 involves the structural support of the nucleus,chromatin regulation and DNA replication,cell cycle regulation,cell ageing,DNA damage,tissue development and nuclear stiffness regulation.LMNB2 mutation can lead to various diseases,such as lipodystrophy,epilepsy and,ataxia,microcephaly.According to existing studies,LMNB2 is related to the biological properties of colorectal cancer,liver cancer,oesophageal cancer,triplenegative breast cancer,ovarian cancer,lung adenocarcinoma and other cancers.Immunohistochemistry(IHC)and Western blot(WB)detection utilize to explore the clinical significance and potential value of LMNB2 protein expression in breast cancer through bioinformatics analysis and immunohistochemistry(IHC).By analyzing the difference between the expression of LMNB2 in breast cancer tissue and adjacent normal breast tissue and the related clinical parameters of breast cancer.At the same time,we also discussed the role of LMNB2 expression in the development of breast cancer.Methods1.This study used Sangerbox(http://sangerbox.com/)online data analysis platform to analyze the correlation of LMNB2 in breast cancer and adjacent tissues and some clinical parameters of breast cancer from the perspective of gene expression level to explore its significance in breast cancer.2.Archive paraffin specimens of breast cancer and adjacent tissues of 101 patients undergoing surgical treatment in the Department of Breastmilk Surgery of Taizhou People’s hospital from January 2018 to December 2020 were selected and collected,and the IHC experiment detected LMNB2.In addition,six fresh samples from October 2022 to December 2022 were collected,which were required to remove the rapidly frozen tissue during the operation,take the cancer tissue and adjacent normal tissue from the remaining tissue,with the size of 0.5x0.5x0.3cm,and detect LMNB2 by WB experiment.All the above specimens were pathologically diagnosed with breast cancer and corresponding para-cancer tissues.3.Statistical software SPSS25.0 was used to analyze IHC experimental results.The WB test results were analyzed using Photoshop,Image J and GraphPad Prism 9.Results1.The expression of LMNB2 in breast cancer and normal breast tissues was analysed using the Sangerbox online platform.LMNB2 expression differed significantly between breast cancer and normal breast tissues and was significantly higher in breast cancer tissues than in normal breast tissues(P< 0.05).The expression of LMNB2 in breast cancer tissues correlated with tumour size(P<0.05)but not with clinical stage,gender,age at onset,lymph node metastasis,and whether distant metastasis was present(P > 0.05);it correlated with breast cancer prognostic parameters OS,PFI,DFI,and DSS(P < 0.05).2.The results of the IHC experimental study showed that LMNB2 expressed to different degrees in breast cancer and normal paraneoplastic tissues.The expression in the former was higher than that in the latter.It could calculate that the high expression rate in breast cancer was significantly higher than in normal paraneoplastic tissues.This result was consistent with the raw letter analysis of LMNB2 m RNA in breast cancer tissues paraneoplastic normal tissues and was statistically significant(P < 0.05).3.Experimental IHC studies showed that LMNB2 expression was not significantly associated with age size,clinical TNM stage,or neural invasion and skin invasion in breast cancer patients(P > 0.05).It was associated with tumour size,whether lymph node metastasis or vascular metastasis(P < 0.05).4.Experimental IHC studies showed that LMNB2 expression was not significantly(P > 0.05)related to PR,Ki-67 index and histological grading.LMNB2 expression correlated with HER-2 and ER expression(P < 0.05)and is negatively correlated with its expression of HER-2 receptor(P < 0.05)and positively correlated with ER receptor(P < 0.05),but the correlation was weak.5.The WB experiments showed that LMNB2 expresses in breast cancer and normal breast tissues,and the difference between the two results was statistically significant(P < 0.05).If the two compare,the expression of LMNB2 in breast cancer tissues was higher than that in control normal breast tissues.ConclusionsIn this study,we found that LMNB2 protein expression was increased in breast cancer,and its high expression was associated with tumor size,whether lymph node metastasis,vascular metastasis,and expression of HER-2 and ER receptors,and correlated with breast cancer prognostic parameters OS,PFI,DFI,and DSS;and it was negatively correlated with HER-2 receptor expression and positively correlated with ER receptor expression.High LMNB2 expression was associated with breast cancer It is inferred that it plays an important role in breast cancer by correlating with some biological behaviors of breast cancer.
