objectiveBy detecting proliferation of rat aortic smooth muscle cell which was transfected Egr-1 decoy oligonucleotide and Egr-1 and PDGF-A expression, detection proliferation of vascular smooth muscle cell, expression changes of early growth response factor-1 and platelet derived growth factor-A (PDGF),to approach the effect and mechanism of the decoy olligonucleotide of the early growth response factor-1 on the proliferation of vascular smooth muscle cells(smc) of rats in vivo.MethodAfter the culture of original arterial SMC form rats with tissue explants adherent method, Design and Synthesis of Egr-1 decoy to lure control oligonucleotides, on vascular smooth muscle cells Immunohistochemistry identification. Transfected synthetic against Egr-1 decoy oligonucleotide (Egr-1 decoy ODN).Packet the cultured cells into normal control group (normal cultured cells), decoy group (transfection Egr-1 decoy oligonucleotide), lured to the control group (Egr-1 transfection decoy oligonucleotide miscellaneous yards).Transfected synthetic against Egr-1 decoy oligonucleotide (Egr-1 decoy ODN)exam the changes of smcs' proliferation and the exression of egr-1 and platelet derived growth factor-A (PDGF-A),which is its downstream related gene using immunohistochemical method.All data used SAS8.0 statistical software for statistical analysis, measurement data to the mean±standard deviation (X±S) said that the comparison group of single-factor analysis of variance,P <0.05 with statistical significance.ResultsImmunohistochemical identification of the cell, identified as high-purity cultured cells in vascular smooth muscle cells. For transfection of Egr-1 lured by immunohistochemical analysis expression of Egr-1 and platelet derived growth factor-A(PDGF-A), cell immunohistochemistry analysis showed that:Egr-1 expression at the three group.Compared with normal control group, decoy oligonucleotide transfection, the expression of Egr-1 and PDGF-Ais be inhibited.ConclusionThis experiment confirm Egr-1 is an important nuclear transcribe factor.can promote vascular smooth muscle cells proliferation.transfection the decoy oligonucleotide of egr-1 can inhibit the proliferation of the arterial smcs of rat in vitro,reduction the exression of egr-1 and PDGF, Achieve the purpose of inhibiting cell proliferation. Further proof of Egr-1 in the anti-atherosclerosis and vascular restenosis treatment plays an important role, so as to further narrow gene therapy has opened up a new way.
