Objective: To explore the different parts of extracts of Shuganhewei Decoction on liver stagnation and spleen deficiency of functional gastrointestinal adverse weight rat,gastrointestinal motility and serum neurotransmitter, protein expression in gastric tissue,screening out the treatment of liver stagnation and spleen deficiency in rats with functional dyspepsia better effective components extraction.Methods: The extraction of petroleum ether extract of liver and stomach soup Decoction by extraction principle, ethyl acetate, n-butanol extract, raffinate by clamping the tail parts. Irregular diet drink acid synthesis method made of liver stagnation and spleen deficiency syndrome of functional dyspepsia. The animal model of agent was determined by enzyme linked immunosorbent (ELISA) treatment in rat serum after serum GAS,MTL,CCK transmitter,AChE,SP,5-HT,ET-1,NO,and Western respectively.The contents of Blot and immunohistochemical determination of Ghrelin protein in gastric antrum of rats, the expression of 5-HT protein.Results:1.The effect of Shuganhewei Decoction extracts on functional dyspepsia rat gastrointestinal motility.Gastric residual rate: normal control group,n-butanol group,ethyl acetate group,liquid group, mosapride group and model group compared group (P < 0.01); group and n-butanol the normal group (P < 0.05); n-butanol group, ethyl acetate group, stoste group and Mosapride group (P>0.05). The Rate of small intestine: normal control group,group, n-butanol, ethyl acetate, group, liquid group, mosapride group and model group, compared with the residual liquid group (P<0.01); n-butanol group and the normal group (P < 0.05); n-butanol group, ethyl acetate group, liquid group and Mosapride group (P>0.05).2.The effect of n-butanol and ethyl acetate on the functional dyspepsia rats(1)After the end of Administration (1), the rats weight and the comparison group(P<0.01), (II) and (VI) comparison (P < 0.01),IV and VI comparison (P<0.05),the difference was statistically significant(2)Gastrointestinal motility: Compared with I, the residual rate of stomach in group VI was higher (P <0.05), and the small intestine propulsion rate was lower in group ? (P<0.01) (P <0.05). Compared with group ?, the residual rate of stomach in group ?, ?and ? was significantly lower than that in group VI (P <0.01), and group III (P<0.05).The small intestine propulsion rate in group II was significantly higher than that in group? (P <0.05), and the small intestine propulsion rate in group ?,? and ? was higher than that in group ? (P <0.05). The rate of intestine in group III was significantly lower than that in group ? (P <0.05), and the rate of small intestine was significantly lower (P<0.05) There was no significant difference in the residual rate and intestine rate between the two groups (P>0.05).(3) Serum neurotransmitter: CCK: n-butanol group and ethyl acetate solution group, model group, control group with significant difference (P < 0.01), n-butanol group and normal control group and Mosapride group showed no significant difference.GAS: the normal group and the ethyl acetate group had significant difference (P< 0.01), n-butanol group and normal group and Mosapride group showed no significant difference (P>0.05) between.MTL: mosapride group and n-butanol group (P>0.05);comparison of ethyl acetate and n-butanol group (P < 0.01).AChE: n-butanol group and Mosapride group (P> 0.05). Comparison of liquid group and n-butanol group (P<0.05).(4) The expression of Ghrelin protein and 5-HT protein:?the expression of Ghrelin protein: n-butanol group and Mosapride group with the deepest color, color liquid group,ethyl acetate group and control group were higher than the normal group the color expression of shallow;?5-HT protein: normal group, n-butanol group, mosapride group was significantly higher than the model control group and the ethyl acetate group.3 the effect of different concentrations of Shugan Hewei decoction n-butanol extract on functional dyspepsia due to stagnation of liver qi and spleen deficiency syndrome(1) WeightTo compare the body weight of the rats after the treatment: n-butanol in low concentration group (M3 group), model group (M1 group) and normal control group(group Z) comparison (P<0.01), the difference was statistically significant. The n-butanol in low concentration group (M3 group), model group (group M1) and Mosapride mosapride group (M2 group) (P<0.01). Compared with Mosapride group (M2 group),normal butanol concentration group (group M4), n-butanol in high concentration group(M5 group) and normal control group (group Z) compare (P > 0.01), there was no significant difference.(2) Gastrointestinal motilityResidual rate of gastric mucosa in each group was significantly higher than that in the control group (M group) and the concentration of n-butanol (M4 group) in the group of moxapril group (M2 group) and n-butanol low concentration group (M3 group) (Group M), but there was no significant difference between the n-butanol concentration group and the n-butanol concentration group (M5 group) (P <0.05), and the difference was significant in the group (P <0.05) .Compared with the group of n-butanol in the low concentration group (M3 group) and n-butanol concentration group (M4 group (P <0.05).There was no significant difference between the two groups (P <0.05), but there was no significant difference between the two groups (M5 group) and the mosapride group (M group) and n-butanol in the low concentration group (M3 group) and n-butanol concentration group (M group) were significantly higher than those in the control group (P<0.01), and the effect of small intestine (P <0.01), but the effect of n-butanol low concentration group (M3 group) and n-butanol concentration group (M4 group) on small intestine propulsion was significantly higher than that of n-butanol group (M5 group)Butanol high concentration group (M5 group) difference.(3) Serum neurotransmittersThe levels of SP,5-HT,ET-1 and NO in the serum of each group were significantly lower than those in the control group (P <0.01). M3,M4 and M5 (P <0.01),there was significant difference, SP content less than M5 group. 5-HT: M3, M4 and M2 groups(P <0.01),there was significant difference, 5-HT content than the Mosapride group less;M5 group and M2 group, there were significant differences, 5- (P <0.05), there was significant difference between M3, M4 group and M5 group, and the content of 5-HT was more than that of mosapride group. ET-1: M1 and Z group,M2, M3, M4,M5 group were significantly different (P <0.01). (P <0.01). There was a significant difference between the M2 group and the M5 group (P <0.01). There was significant difference between the M2 group and the M group (P <0.01), the ET- (P <0.05). There was significant difference between M3 group, M4 group and M5 group. The content of ET-1 in M5 group was higher than that in M3 group and M4 group. NO: M3 group and M2 group(P <0.01), there was significant difference, M3 group NO content is higher than M2 group content. M5 group and M2 group (P <0.01), there was significant difference, M5 group NO content was significantly lower than the M2 group content.(4)Expression of Ghrelin protein in gastric antrum: the expression of Ghrelin protein in the normal control group (group Z) and Mosapride group (M2) the band color gray is similar to that of M3,M4,M1 group were lower than those of Z and M2 color light, the color of M5 most.4 Conclusion(1) N-butanol Shuganheweitang, ethyl acetate, liquid can increase liver stagnation and spleen deficiency syndrome of functional dyspepsia rats weight and long propulsion and gastric emptying, the residual liquid part of almost no increase of liver stagnation and spleen deficiency syndrome of functional dyspepsia long promote gastric emptying. The petroleum ether fraction almost no extraction rate, so the exclusion of petroleum ether and residual liquid extracts of Shuganhewei decoction, screened n-butanol treatment of liver stagnation and spleen deficiency syndrome of functional dyspepsia, extract of ethyl acetate was prepared after the study.(2) According to the preliminary screening of extracts of Shuganhewei decoction,n-butanol extracts and ethyl acetate extracts of gastrointestinal motility on body weight,liver stagnation and spleen deficiency syndrome of functional dyspepsia in SD rats, the expression of serum proteins and neurotransmitters, screened the n-butanol extract for the better parts.(3) To investigate the concentration of 60mg - kg-1, 120mg -kg-1, 240mg - kg-1 Shuganheweitang n-butanol extract on liver stagnation and spleen deficiency of functional gastrointestinal weight, adverse gastrointestinal motility in rats, serum neurotransmitter and protein expression, found that the concentration of 240mg - kg-1 Shuganheweitang n-butanol extract of the best liver stagnation and spleen deficiency syndrome of functional dyspepsia treatment. The study laid the foundation for Shuganheweitang dosage forms the following it.
