[Objective] To investigate effects and anti-free radical mechanism of Polygala on anti-hypoxia in mice.[Methods] Forty mice were randomly divided into four groups, control group, Polygala low dose group(0.3g/kg),middle dose group(0.6g/kg) and large dose group(1.2g/kg). It is ten in each group. The control group was administrated by physiological saline. The anti-hypoxia time was measured after intragastric administration three weeks. The blood was obtained by stabing heart in mice. Superoxide dismutase activity and malondialdehyde content were determined by centrifuging the blood.[Results] 1. Effects of Polygala on common condition and weight in mice:After intragastric administration three weeks with Polygala in mice, the common contion were good. Moreover, all Polygala groups's mice weight hadn't significantly changed compaired with control group (P>0.05).2. Effects of Polygala on anti-hypoxia time:Polygala with middle dose and large dose could increase anti-hypoxia time compared with control group in mice (P<0.05), but not in low dose group(P>0.05).3. Effects of Polygala on SOD and MDA:Polygala with middle dose and large dose could increase SOD and degrade MDA compaired with control group (P<0.05), but not in low dose group(P>0.05).[Conclusion] Polygala could enhance the ability of anti-hypoxia in mice. Its mechanism of action maybe concerned with enhancing anti-free radical in blood serum.
