Fatigue,as a non-specific symptom,exists in healthy or diseased human beings,especially when the exhausted body cannot be recovered after overtraining,it will evolve into fatigue syndrome,and even cause health threats to the immune system,endocrine system,central nervous system,etc.More than 20% of the population is suffering from fatigue,and one third of them are actively seeking help from medical institutions.Polygala tenuifolia wild.,also known as "Xiaocao",is a dry aerial parts of Polygala tenuifolia wild..It has the effect of "tonifying essence,nourishing Yin Qi,deficiency damage and dream discharge".Modern research shows that Polygala Herb has good antioxidant and anti-aging activities.Therefore,in this study,the anti-fatigue and hypoxia tolerance effects of Polygala Herb extract were studied,and the main research contents and results were as follows:1.The anti-fatigue and hypoxia tolerance activities of different aerial parts of Polygala tenuifolia wild.extract(APT)were evaluated by weight-loaded swimming test,hypoxia tolerance under normal pressure and sodium nitrite poisoning survival test.Compared with the control group,the different APT groups can prolong the time of weight-loaded swimming,the time of hypoxia tolerance under normal pressure and the survival time of sodium nitrite poisoning in mice,of which the effect of 50% alcohol APT group is the most significant,indicating that the APT can alleviate the fatigue and anorexic of mice.Compared with the control group,the APT can reduce the level of serum CK,BUN and LA,clear the accumulation of physical exercise metabolites,improve the level of SOD and GSH in serum and liver tissue,reduce the level of MDA,enhance the activity of antioxidant enzymes,prolong the performance of exercise,and reduce the occurrence of physical fatigue and hypoxia.2.The anti-fatigue effect and mechanism of 50% alcohol APT were evaluated by weight-loaded swimming test and fatigue rotating rod test in mice.The 50% alcohol APT can significantly improve the time of weight-loaded swimming and rotating rod in mice.Compared with Rhodiola capsule group(Rho),the effect of middle and high dose groups on the time of weight-loaded swimming is more obvious.APT group significantly reduced the accumulation of sports metabolites,increased the activity of antioxidant kinase,decreased the level of lipid peroxide,and increased the level of ATP and SDH in mice liver muscle.In addition,APT showed no side effects on the clinical biochemical indexes and pathological sections of liver and kidney function in mice.3.The mice model of kidney-yang deficiency was made by hydrocortisone.The anti-fatigue effect and its mechanism were studied by weight-loaded swimming test and fatigue rotating rod test.The results of serum hormone index,pathological section of kidney and testis showed that the endocrine system dysfunction,gonadal axis and thyroid axis dysfunction of the mice showed that hydrocortisone was successful in the preparation of kidney-yang deficiency mice model.The results of weight-loaded swimming test and fatigue rotating rod test showed that the APT can significantly improve the endurance exercise time of kidney-yang deficiency mice,and the results of middle and high dose groups showed that the effect was better than that of Shenrong Bushen(SRBS)tablet group.Compared with the model group,the group of APT reduced the levels of serum CK,BUN,LA,SOD,GSH,MDA,ATP,SDH and MDH.Western blot was used to detect the protein expression of AMPK and Nrf-2 in the liver of mice.The results showed that APT could improve the protein expression of AMPK and Nrf-2.
