The Expression And Clinical Significance Of Survivin,VEGF,Bcl-2 And Ki67 In Colorectal Cancer

Objective:Colorectal cancer(CRC) is one of the most common cancers.The incidence and development of colorectal cancer are related to many factors,including a variety of the cancer gene activation and the cancer suppressor gene deactivation which play an important role in this process. To analyze the expression and clinical significance of Survivin,VEGF,Bcl-2 and Ki67 in colorectal cancer by detecting their expression in cancerous and adjacent tissues(more than 5cm from the cancerous tissue) in this study.Methods:Example of 72 cases were selected at random from colorectal cancer patients who were treated in tumor hospital of Liaoning province from December 2009 to February 2010, Male 41, Female 31, aged from 28 to 77 years old, and the mean age was 58. Age≥60 years old 33, Age<60 years old 39. All selected patients had not received adjuvant chemotherapy or radiotherapy before operation, and had no secondary operation history. Get each part respectively in cancerous and adjacent tissues (>5cm from the cancerous tissue). The expression of Survivin,VEGF,Bcl-2 and Ki67 in all cases were detected by immune histochemistry, and the data were examined by chi-square test. P<0.05 was considered statistic significance.Results:Survivin was mainly localized in the nucleus or cytoplasm of colorectal cancer, with performance of yellowish brown pellet and staining of Heterogeneity. The positive expression rate of Survivin in cancerous tissues was 68.1%(49/72), and the adjacent tissue was negative. The appearance of yellowish brown pellets in cytoplasm was the positive reaction of VEGF. The positive expression of VEGF in this group of 72 cases of cancerous tissues was 62.5% (45/72), and that of adjacent tissues was 20.8%(15/72). The positive expression of Bcl-2 in this group of cancerous tissues was 55.6% (40/72), and adjacent tissues was 15.3% (11/72), with performance of yellowish brown pellet in cytoplasm and cell membrane, distribution of focus or diffuse. The cell nucleus present yellow or yellowish brown was the positive reaction of Ki67. The positive rate of Ki67 in cancerous tissues was 84.7% (61/72), and that in adjacent tissues was 30.6% (22/72). The statistic analysis demonstrated that the positive expression of Survivin had significant differences with the tissue differential degree of cancer,CEA,metastasis of lymph mode and Ducks staging; but had no significant differences with sex,age,tumor position,tumor size and the General classification of tumor. The positive expression of VEGF had significant differences with tumor size,CEA,the tissue differetial degree of cancer,metastasis of lymph mode and Ducks staging; but had no significant differences with sex,age,tumor position and the General classification of tumor. The positive expression of Bcl-2 had significant differences with the tissue differential degree of cancer,metastasis of lymph mode and Ducks staging; but had no significant differences with sex,age,tumor position,tumor size,CEA and the General classification of tumor. The expression of Ki67 had no significant differences with the clinical index of colorectal cancer. The differences of these four gene expression between cancerous and adjacent tissues were significant.Conclusion:1. The positive expression of Survivin had relationship with the tissue differential degree of cancer,CEA,metastasis of lymph mode and Ducks staging; but had no relationship with sex,age,tumor position,tumor size and the General classification of tumor.2. The positive expression of VEGF had relationship with tumor size,CEA,the tissue differential degree of cancer,metastasis of lymph mode and Ducks staging; but had no relationship with sex,age,tumor position and the General classification of tumor.3. The positive expression of Bcl-2 had relationship with the tissue differential degree of cancer metastasis of lymph mode and Ducks staging; but had no relationship with sex,age,tumor position,tumor size,CEA and the General classification of tumor.4. The expression of Ki67 had no relationship with the clinical index of colorectal cancer.5. The differences of these four gene expression between cancerous and adjacent tissues were significant.
