Objective:To explore the role of IGF-1、IGF-1R and IGFBP-2in the occurrence anddevelopment of human colorectal carcinoma by detecting the expression ofthese factors in normal mucosa, colorectal adenoma and colorectal carcinomaas well as the relationship among factors and the relationship of these factorswith the clinical pathological characteristic of colorectal cancer patients suchas gender, age, differentiation, depth of invasion, lymph node metastasis, andDuke’s stage so as to evaluate the relationship between these factors andprongosis.Method:50cases of colorectal carcinoma (CC),20cases of colorectal adenoma(Ad) and20cases of colorectal normal mucosa(NM)that were surgicallyremoved and certified by pathology from July,2012to March,2013, fromSurgical Oncology of Chengde Medical College affiliated hospital werecollected. RT-PCR was used to examine the mRNA of IGF-1、IGF-1R andIGFBP-2,immunohistochemical staining was used to examine expression ofIGF-1、IGF-1R and IGFBP-2.Results:1The expression of IGF-1mRNA, IGF-1R mRNA, IGFBP-2mRNA, IGF-1、IGF-1R and IGFBP-2in normal mucosa, colorectal adenoma and colorectalcarcinoma.1.1The expression of IGF-1mRNA, IGF-1R mRNA and IGFBP-2mRNA inNM, Ad, CCIGF-1mRNA in CC was significantly higher than that in Ad (P<0.01) and NM (P<0.01). Also IGF-1mRNA in Ad was signifycantly higher than thatin NM (P<0.01).IGF-1R mRNA in CC was significantly higher than that in Ad (P<0.01)and NM (P<0.01). Also IGF-1R mRNA in Ad was signifycantly higher thanthat in NM (P<0.01)IGFBP-2mRNA in CC was significantly higher than that in Ad (P<0.01)and NM (P<0.01). Also IGFBP-2mRNA in Ad was signifycantly higher thanthat in NM (P<0.01)1.2The expression of IGF-1、IGF-1R and IGFBP-2in NM, Ad, CC1.2.1The expression of IGF-1in NM, Ad, CCThe expression of IGF-1in CC was significantly higher than those of inAd (P<0.01) and NM (P<0.01). Also the expression of IGF-1in Ad wassignificantly higher than those of in NM (P<0.01).1.2.2The expression of IGF-1R in NM, Ad, CCThe expression of IGF-1R in CC were significantly higher than those ofin Ad (P<0.01) and NM (P<0.01). Also the expression of these two factors inAd were significantly higher than those of in NM (P<0.01).1.2.3The expression of IGFBP-2in NM, Ad, CCThe expression of IGFBP-2in CC were significantly higher than those ofin Ad (P<0.01) and NM (P<0.01). Also the expression of these two factors inAd were significantly higher than those of in NM (P<0.01).2The relationship between IGF-1mRNA, IGF-1R mRNA, IGFBP-2mRNA,IGF-1、 IGF-1R and IGFBP-2and clinical pathological characteristic ofcolorectal carcinoma.2.1The relationship between IGF-1mRNA, IGF-1and pathologicalcharacteristic of CCThe expression of IGF-1mRNA and IGF-1were positively correlatedwith depth of invasion, lymph node metastasis, Duke’s stage. The expressionof IGF-1mRNA, IGF-1of infiltration more than musucularis in colorectalcarcinoma was higher than not (P<0.05, P<0.05).The positive expression ofIGF-1of lymph node metastasis group was significantly higher than those without lymph nodemetastasis group (P<0.01, P<0.01).The positiveexpression of IGF-1of Duke’s CD stage was significantly higher than Duke’sAB stage,(P<0.01, P<0.01).Correlations were not found between IGF-1mRNA, IGF-1and everyother characteristics of CC such as gender, age, differentiationdegree,(P>0.05,P>0.05,P>0.05).2.2The relationship between IGF-1R mRNA, IGF-1R and pathologicalcharacteristic of CCThe expression of IGF-1R mRNA and IGF-1R were positively correlatedwith depth of invasion, lymph node metastasis, Duke’s stage. The expressionof IGF-1R mRNA, IGF-1R of infiltration more than musucularis in colorectalcarcinoma was higher than not (P<0.05, P<0.05).The positive expression ofIGF-1R of lymph node metastasis group was significantly higher than thosewithout lymph nodemetastasis group (P<0.01, P<0.01).The positiveexpression of IGF-1R of Duke’s CD stage was significantly higher than Duke’sAB stage,(P<0.01, P<0.01).Correlations were not found between IGF-1R mRNA, IGF-1R and everyother characteristics of CC such as gender, age, differentiationdegree,(P>0.05,P>0.05,P>0.05).2.3The relationship between IGFBP-2mRNA, IGFBP-2and pathologicalcharacteristic of CCThe expression of IGFBP-2mRNA and IGFBP-2were positivelycorrelated with depth of invasion, lymph node metastasis, Duke’s stage. Theexpression of IGFBP-2mRNA, IGFBP-2of infiltration more than musucularisin colorectal carcinoma was higher than not (P<0.01, P<0.05).The positiveexpression of IGFBP-2of lymph node metastasis group was significantlyhigher than those without lymph nodemetastasis group (P<0.01, P<0.01).Thepositive expression of IGFBP-2of Duke’s CD stage was significantly higherthan Duke’s AB stage,(P<0.01, P<0.01).Correlations were not found between IGFBP-2mRNA, IGFBP-2andevery other characteristics of CC such as gender, age, differentiation degree,(P>0.05,P>0.05,P>0.05).3The relationship among IGF-1、IGF-1R and IGFBP-23.1The relationship between IGF-1and IGF-1RThe positive relationship could be seen between IGF-1andIGF-1R(r=0.585,P<0.01).3.2The relationship between IGF-1and IGFBP-2The positive relationship could be seen between IGF-1andIGFBP-2(r=0.629, P<0.01).3.3The relationship between IGF-1R and IGFBP-2The positive relationship could be seen between IGF-1R andIGFBP-2(r=0.603, P<0.01).Conclusin:1The high expression of IGF-1、IGF-1R and IGFBP-2may play an importantrole in the occurrence and development of colorectal cancer.2IGF-1、IGF-1R and IGFBP-2was significantly correlated with depth ofinvasion,lymph node metastasis and Duke’s stage in colorectal carcinoma. Sothey could be a predictive and prognostic factor of CC.3There were significantly positive correlation among IGF-1、IGF-1R andIGFBP-2in colorectal carcinoma.4It can be predicted by some way to inhibit or block the expression of IGF-1、IGF-1R and IGFBP-2or one of them, May reduce the invasive ability ofoverexpressed factors and colorectal cancer, so as to reduce the probability ofmetastasis in colorectal carcinoma.
