Objective:To investigate the clinical significance of MMP-2 and TIMP-2 in normal cervix, cervical intraepithelial neoplasia (CIN) and cervical squamous cell carcinoma, and the relationship in invasion and metastasis of cervical squamous cell carcinoma between MMP-2 and TIMP-2Methods:Powervision immunohistochemical method was employed to detect the expression of MMP-2 and TIMP-2 in 30 cases of normal cervical,48 cases of CIN and 50 cases of cervical squamous cell carcinoma.The statistical software-SPSS 17.0 was applied to analyze the results.Results:1. The positive rates of MMP-2 expression in normal cervix, CIN and cervical squamous cell carcinoma were 6.66%,37.50%,70.00%, three statistically significant difference (P<0.05); The positive rates of TIMP-2 expression in normal cervical, CIN and cervical squamous cell carcinoma were 10.00%,20.83%,32.00%, no significant difference between the three groups(P>0.05); but the positive rates of TIMP-2 expression in normal cervix and cervical squamous cell carcinoma of the difference was statistically significant (P<0.05). 2. The positive rates of MMP-2 expression in CINⅠ, CINⅡ, CINⅢwere 15.38%,29.41%,61.11%, three statistically significant difference (P<0.05). The positive rates of TIMP-2 expression in CINⅠ, CINⅡ, CINⅢwere 7.69%, 17.65%,33.33%, no significant difference between the three groups (P> 0.05).3. In cervical squamous cell carcinoma, the positive rates of MMP-2 expression in differentiation asⅠ-level,Ⅱ-level,Ⅲ-level were 30.00%,70.58%, 86.96%, three statistically significant difference (P<0.05); at stagesⅠ-Ⅱa period (57.14%,)was lower than theⅡb-Ⅳperiod(86.36%) (P<0.05); there is lymph node metastasis group the positive rate of 90.00%, higher than the non-lymph node metastasis in 42.86%, the difference was statistically significant (P<0.05). Expression of TIMP-2 in cervical squamous cell was no related with histological types (P>0.05); at stagesⅠ-Ⅱa period(46.43%) was higher than theⅡb-Ⅳperiod (13.64%)(P<0.05); there is lymph node metastasis positive rate was 10.00% lower than those without lymph node metastasis of 57.14%, the difference was statistically significant (P<0.05).4. MMP-2 and TIMP-2 expression in cervical squamous cell carcinoma showed negative correlation.Conclusion:1. The positive rates of MMP-2 expression in normal cervix, CIN, cervical squamous cell carcinoma was gradually increased, MMP-2 may be involved in the occurrence and development of cervical squamous cell carcinoma.2. The positive rates of MMP-2 expression in cervical squamous cell carcinoma was positively correlated with degree of differentiation, clinical stage and lymph node metastasis;while the expression of TIMP-2 was negative related, it suggested that both can be used as predictable prognosis of patients with cervical squamous cell carcinoma of reference.3. MMP-2 and TIMP-2 in cervical squamous cell carcinoma there is a negative correlation between the expression, it suggested that both play in the development of cervical cancer antagonism.
