Objective: To investigate the expressions of SIRT-1 protein in different cervical tissues(low-grade intraepithelial neoplasia and high-grade intraepithelial neoplasia)and it clinical significance.Method: From october,2014 to january,2016,we collected tissue samples for surgical treatment or cervical biopsy of 140 patients,including 26 cases of LSIL? 35 cases of HSIL and 55 cases of cervical cancer,and there were 24 cases of normal cervix that Surgical treatment for uterine fibroids,comes from the Affiliated Hospital of Medical College Qingdao University gynaecology and Qingdao Women and Children's Hospital gynaecology center.Applying immunohistochemical method(SP)and western-blot,were detected for SIRT-1 protein and their correlation with clinicopathological features of the cancer was analyzed.Result: 1.Immunohistochemistry showed that SIRT-1 protein was expressed mainly in the nucleus,and the staining of brown granules was positive(Figure 1).The positive expression rate of SIRT-1 protein in normal cervical tissues,LSIL,HSIL and cervical cancer tissues were 0,15.38%,37.14%,40.00%,the differences among tissues being significant(X2 = 16.82,P < 0.05).2.Western-blot showed that SIRT-1 protein was same to the Immunohistochemistry,compared significantly the difference between the four groups was statistically significant(F=13.86,P<0.05)(Figure 3).3.The expressions of SIRT-1 protein in cervical cancer tissue were closely associated with the degree of differentiation,with or without lymph node metastasis and FIGO staging,the lower the differentiation degree,the higher the positive expression rate(X2=6.87,P<0.05);The positive expression rate of lymph node metastasis positive group was higher than that of lymph node metastasis negative group,the difference was statistically significant,(X2=7.73,P<0.05);the positive rate of SIRT-1 protein increased with the increase of clinical stage,the difference was significant(X2=7.39,P<0.05).4.And that in cervical cancer tissue was not significantly correlated with the pathological type and the age of the patients and the difference is not statistically significant(X2=1.03?0.038,P>0.05).Conclusion:1.The expression of SIRT-1 protein in normal cervical tissue?LSIL?HSIL?cervical cancer tissue gradually increased,suggesting that the gradual process of the occurrence and development of SIRT-1 may be involved in the normal cervical tissue and low grade intraepithelial lesions-high-grade intraepithelial lesions of cervical cancer.2.With the high expression of SIRT-1 protein in pathological lower differentiation,lymph node metastasis,clinical stage of cancer patients,suggesting that the abnormal expression of SIRT-1 protein in cervical cancer may play an important role in the occurrence,development and metastasis.3.SIRT-1 protein is expected to become a biological marker of early diagnosis and prognosis of cervical cancer,and provide a new target and scientific evidence for the prevention and control strategy of cervical cancer.
