Acanthopanax senticosus is known for thousands of years as a medicinal herb. It has been used to treat neurosis clinically, but the mechanism of its anti-stress is not very clearly. The protective effect of Acanthopanax senticosus extract (ASE) on effect on the behaviors and neurotransmitter of anxiety and depression mice, PC12 injured by caffeine, effect on the serum enzyme of the cold stress chicken are studied to elaborate the anti-stress mechanism of ASE.1. This study utilized mice subjected to six universal stress tests (the elevated plus maze, the light-dark transition test, the tail suspension test, the forced swimming test, the hot plate test, the traction test) to study the effect of ASE on resistance to stress. The behaviors of the model mice were detected.The results showed that 1000 mg/kg ASE significantly increased the proportion of entries in the open arm during the elevated plus maze and the total time standing in the light room during the light-dark transition test; In addition,500~2000 mg/kg ASE reduced the duration of immobility during the forced swim test, while 2000 mg/kg ASE reduced the duration of immobility during the tail suspension test; 500~1000 mg/kg ASE had no significant effect on the pain threshold; 500~2000 mg/kg ASE had no significant effect on the traction.The mice tests showed that ASE had effect of anti-anxiety and anti-depressant without side effect such as analgesic and muscle relaxation.2. We examined the effects of ASE on the concentration neurotransmitter (dopamine, 5-hydroxy tryptamine and noradrenalin) in the mice cerebrum and the cell viability of PC 12 injured by caffeine to study the mechanism of anti-anxiety and anti-depressant of ASE.During the tail suspension test, the concentration of 5-hydroxy tryptamine was increased by 2000 mg/kg ASE. The cell viability could be increased by 12~24 mg/kg ASE. The study suggest that ASE has the effect of increased the concentration of 5-hydroxy tryptamine and reducing the excitotoxicity by caffeine, which resulte the effect of anti-anxiety and anti-depressant.3. Cold stress chicken were fed diet supplement with 2% ASE, the activities of serum enzyme (AST,ALT,LDH,CK,y-GT) and survival rate were detected. The results showed that the activities of AST,ALT,y-GT were reduced, the activities of CK were increased by ASE, but there were no significant effect on LDH. The survival rate was increased by ASE during the acutecold stress. It means that ASE may reflect the anti-cold stress effect by adjusting the activities of serum enzyme.It could be concluded that Acanthopanax senticosus extract have the effects of anti-anxiety, anti-depressant, anti-cold stress, and the effects are associated with its ability of protective effects of neuronal, adjusting theconcentration of 5-hydroxy tryptamine and activities of serum enzyme.
