Acanthopanax senticosus Harm.has been used as a traditional oriental medicine with tonic,antirheumatic,longitudinal bone growth,adaptogenic,antifatigue,antitumor and gastric ulcer activity,which possesses the prophylactic functions for chronic bronchitis hypertension,antistress,ischemic heart disease.Clinical experience and pharmacodynamic experiments have showed that the root of Acanthopanax senticosus Harm.extract has antifatigue effect.But the active substantial basis and function principle of the root extract have not been elucidated until now.Antifatigue effects of As1 and As2 were evaluated by mice burden swimming testing. As1 showed an excellent antifatigue activity,but,the effect of As2 was lower than that of As1.The result showed that both As1 and As2 possessed antifatigue activity,suggesting that As1 was one of the major active fractions of Acanthopanax senticosus Harm..Compounds 1-8 were isolated from the antifatigue fractions(As1 and As2)of Acanthopanax senticosus Harm.,using various column chromatography and semi-preparative HPLC.The structures of the compounds obtained were determined by modern spectroscopic methods(1H-NMR,13C-NMR,HMBC,ESI-MS)as following:7-α-D-glucopyranosyloxy-6, 8-dimethoxy-2H-1-Benzo-payran-2-2one(Eleutheroside B)(1),β-D-glucopyranoside,4-(3-hydroxl-1-propenyl)-2, 6-di-methoxyphenyl(2),bis(2,6-dimethoxy-4,1-phenylene)bis-, [1R-(lα,3aβ,4β,6aα)]-β-D-glucopyranoside-tetrahydro-1H,3H-furo[3,4-c]furan-1, 4-diyl(3),chlorogenic methyl ester(4),chlorogenic acid(5),amygdaloside(6),Daucosterol (7),β-sitosterol(8).A HPLC method had been set up for qualitative and quantitative analysis of the main component(Eleutheroside B)in As1,the major active fraction of Acanthopanax senticosus Harm.,The calibration cure was linear in the range of 0.104-20.80μg(r = 0.999 9),the average recovery was 97.68%,RSD was 1.40%(n = 6).This method was simple,accurate and reliable.The average contents of Eleutheroside B in As1 was 7.45%,RSD was 1.56%.In conclusion,it was proved that As1 and As2 possessed antifatigue activity,providing good bases for its further R&D.Eight compounds were isolated from As1 and As2 and the structures of them were determined by spectral analysis.These compounds,which may be the effective active components for antifatigue from the active site of Acanthopanax senticosus Harm.,may be developed as drugs or as leading compounds.
