| Objectives:To explore the effect of spirit stress events on the formation of atherosclerosis an its drug prevention.Methods:Establish atherosclerosis model by feeding in rabbits with high-cholesterol food and randomly divided them into 6 groups: high-cholesterol with stress controls by sodium ferulate intervention group (Group A,n=10), high-cholesterol with stress controls by simvastatin intervention group(Group B,n=10), high-cholesterol with stress controls by no-intervention group(Group C,n=10), high-cholesterol without stress controls group(Group D,n=10), stress without controls group(Group H,n=10), and blank control group (Groupâ… ,n=10). After 12 week of feeding high-cholesterol food, stimulate rabbits in group A,B,C,H for 7days to build stress model. Draw blood of all animals on dayl, week 12 and week12. Assay TC,TG,LDL,HDL level of blood before diet by BackmaanLX 20 Automatic chemistry Analyzer. Assay ET-1, NE, hs-CRP, IL-6 level in blood with ELISA assay. After 13 weeks, kill all the animals. Select the part of the aorta of the rabbits, oil red "O" staining capture the image with camera and analysis the area of plaque using computer image software. Intercept about 3mm of the aorta, HE staining sections in each group to observe morphology and plaque formation. Intercept the aorta, staining by sodium acetate and citric acid, and observe the plauqe formation by transmission electron microscope. ResultsET-1, NE, hs-CRP, IL-6 levels in groups with stress are significantly higher after stimulation, P<0.05; TC,TG,LDL,HDL levels in groups with high-cholesterol food are higher before, P<0.05。percentage of plaque area are higher in group of high-cholesterol food with stress than the group of high-cholesterol food without stress, P<0.05; percentage of plaque area in simvastatin and sodium ferulate intervention group are lower than the group of high-cholesterol food with stress. P<0.05; and there are no statistical difference among simvastatin intervention group and sodium ferulate intervention group. Pathological:Light microscope, visible palque are observed on the surface of aortic intima in all specimens of high-cholesterol diet groups; collagen-rich foam cells and some calcified plaque necrosis are also observed in this region. Electron microscope, degeneration and necrosis of endothelial cell can be seen in groups with high-cholesterol food and stress group.Conclusion:The psychological stress can induce the significantly increasing of ET-1,NE,hs-CRP,IL-6 in the serum; may directly or indirectly lead to endothelial cell damage and dysfunction; sodium ferulate and simvastatin may act on endothelial cells, antagonism of stress injury, against the stress caused by vascular endothelial dysfunction. |
