Objective To establish the normal full-term newborn reference values, cord blood and urine 8-iso-PGF2a levels in normal neonates with gestation age (GA) 37-41 weeks were determinated. To investigate the possibility for assessing lipid peroxidation via determination of urine 8-iso-PGF2a level, the correlation of cord blood and urine 8-iso-PGF2 a levels in neonates with GA 28-43 weeks was analysized. To estimate lipid peroxidation in the premature and post-term neonates, cord blood and urine 8-iso-PGF2a levels in these neonates were compared with ones in the term neonates.Methods Neonates with GA 28-43 weeks were selected as subjective. According to GA, these neonates were divided into four groups, i. e. Group A:GA 28-32 weeks, 50 cases; Group B:GA 33-36 weeks,62 cases; Group C:GA 37-41 weeks,85 cases; Group D:GA 42-43 weeks,25 cases. The samples of umbilical blood and urine were collected from the neonates in 24 hours after birth. Cord blood and urine 8-iso-PGF2a levels were measured by ELISA.Results①Cord blood 8-iso-PGF2a level from the normal full-term newborn was 130.09±31.73 pg/ml, the 95% confidence interval (CI) was 123.25-136.94pg/ml; urine 8-iso-PGF2a level was 27.14±6.73 pg/ml, the 95% CI was 25.69-28.59pg/ml.②Cord blood 8-iso-PGF2a levels from premature infant were respectively 188.42±59.34 pg/ml (GA 28-32 weeks) and 189.37±68.46pg/ml (GA 33-36 weeks), which had no significant differences (t=0.0744, p=0.9384), but all of these two groups were higher than 37-41 week full-term newborn levels (t=7.4436, p=0.0000; t=7.0221, p=0.0000).③Urine 8-iso-PGF2a levels from premature infant were 32.14±7.32 pg/ml (gestational age 28-32 weeks) and 30.46±8.83 pg/ml (GA 33-36 weeks) which had no significant differences (t=1.0789, p=0.2830), But all of these two groups were higher than 37-41 week full-term newborn levels (t=4.0347, p=0.0001; t=2.5871, p=0.0107).④The cord blood and urine 8-iso-PGF2a levels in normal neonates (GA 28-43 weeks) had direct correlation, the correlation coefficient was 0.7685 (t=18.7224, p=0.0000).Conclusions The reference values of cord blood and urine 8-iso-PGF2 a were estabilshed in normal full-term newborn, and the 95% CI were 123.25-136.94pg/ml and 25.69-28.59pg/ml, respectively. There were lipid peroxidation damage in premature infant and post-term infant, and their serum 8-iso-PG2a level rises along with the illness course. The use of ambroxol (30mg/kg·d) display a good anti-oxidation effect. Urine 8-iso-PGF2a levels can reflects lipid peroxidation in neonates,and its determination had important clinical value.
