Objective To establish a rat model with intermingled transplantation of small stamps of auto-alloskin on full-thickness defect wounds in rats and observe the influence on the wound healing of this grafting method. Explore the methods of tracing the allogeneic skin after this intermingled transplantation.Methods Full-thickness skin defect wounds were made on the back of the rat. Intermingled transplantation of auto-allo small stamps skin on the wounds were performed. The auto-and allo-small stamps skin grafting were performed respectively as the controlled group. The growth of the grafted skin, the wound healing rate, wound contraction rate and histological changs were observed on the 1st.2nd.3rd.4th weeks after grafting.Results All the grafted skin adhered with recipient bed well, became red and alive 1 week post-grafting.The grafted skin in autograft group confluenced gradually. The dry crust in the surface of the allogeneic skin in the intermingled grafting group 2 weeks after grafting while the grafted skin become black, swelled and necrotic progressively in the allograft group. The wounds of auto-and intermingled grafting groups healed well in 4 weeks. But the grafted allogeneic skin disappeared and the granulation tissue formed in the wounds in allograft group.The wound healing rate in intermingled grafting gruoup was higher than that in the allograft group at the end of the 2nd.3rd.4th weeks after grafting (P<0.05) and was the most obvious (P<0.01) in the 3rd weeks. The wounds in auto-and intermingled grafting group healed and that in allograft group fulled with granulation 4 weeks after grafting. There was no difference of healing rate between auto-and intermingled grafting group during the healing process. And there were also no difference among all these groups in wound contraction. Acute host versus graft reaction was occured in allograft group between epidermis and dermis 2 weeks after grafting and this reaction was lighter in the inermingled grafting group.Y chromosome-specific fragment was not found by Y chromosome-specific PCR in allograft group 3 weeks after transplantation while it was still detected in the intermingled grafting group 6 weeks after transplantation. The green fluorescence expression were both disappeared in allo-and intermingled grafting group in one month by using of green fluorescent protein transgenic mice as donors for traceing.Conclusion This established model could replicated clinical practice well and healed the full thickness defect wounds quickly and relieve the host versus graft reaction by using this intermingled grafting.The method of Y chromosome-specific PCR was a good method for tracing the allogenic skin in skin grafting and green fluorescent protein transgenic mice as donors for tracer should be improved because the tracing time was short.
