OBJECTIVE:Detection the expression of Ets-1 and E-cadherin in esophageal squamous cell carcinoma (ESCC), to analyze the relationship between the expression and clinical pathological characteristics; To explore the interrelation of the expression of Ets-1 and E-cadherin; To discusses the relationship between the expression Ets-1 and E-cadherin and neoplasms infiltration and the lymph node metastasis in ESCC and its clinical significance; To find out the effective and objective parameters for anticipation of lymph node metastasis of ESCC,and to provide a new method for carcinoma treatment.METHODS:10 case of adjacent noncancerous tissues and 62 cases of ESCC tissues were choosen to detect the expression of Ets-1 and E-cadherin by immunohistochemical SP staining. Combined with clinical pathological characteristics to analyze using statistical method.RESULTS:(1) The positive expression rate of Ets-1 was 0 (0/10) in 10 case of adjacent noncancerous tissues,64.5% (40/62) in 62 cases of ESCC tissues; From adjacent noncancerous tissues to ESCC tissues, the positive expression rate of Ets-1 was gradually increase. There was statistical difference between them (r= 12.02, P<0.05); The positive expression rate of Ets-1 was 74.4%(29/39) in lymph node metastasis group,47.8% (11/23) in no lymph node metastasis group.There was statistical difference and positively correlation between them (rs=0.396, P<0.05).(2) The positive expression rate of Ets-1 was not related to gender and age of patients, and the size, location of tumor, there was not statistical difference between them (P>0.05, respectively), but there was positively correlated with the TNM staging,degree of differentiation, depth of invasion, there was statistical difference between them(rs>0, P<0.05, respectively).(3) The positive expression rate of E-cadherin was 90%(9/10) in 10 case of adjacent noncancerous tissues,48.4%(30/62) in 62 cases of ESCC tissues; From adjacent noncancerous tissues to ESCC tissues, the positive expression rate of E-cadherin was gradually decrease. There was statistical difference between them (x2= 4.447, P<0.05); The positive expression rate of E-cadherin was 35.9%(14/39)in lymph node metastasis group,69.6%(16/23) in no lymph node metastasis group.There was statistical difference and negatively correlation between them (rs=-0.280, P<0.05)(4) The positive expression rate of E-cadherin was not related to gender and age of patients, and the size, location of tumor, there was not statistical difference between them (P>0.05, respectively), but there was negatively correlated with the TNM staging,degree of differentiation, depth of invasion, there was statistical difference between them(rs<0,P<0.05, respectively).(5) The expression rate of Ets-1 had negatively correlation compared with the expression rate of E-cadherin in ESCC(rs=-0.361,P<0.05).CONCLUSION:(1) The proto-oncogene of Ets-1 was high expression status in ESCC tissues, The positive expression rate of Ets-1 was positively correlated with tumor lymph node metastasis and depth of invasion, it showed that Ets-1 plays a very important role in the invasion and metastasis of ESCC, the detection of Ets-1 can be used as an important index of forecasting tumor metastasis.(2) The expression rate of Ets-1 in ESCC was significantly higher than in adjacent noncancerous tissues, it points out that Ets-1 gene activation may be closely related to occurrence and development of ESCC, blocking synthesis and secretory pathway of Ets-1 can be used as a new means of treatment of ESCC, it help to improve the malignant tumor therapy.(3)The anti-oncogene of E-cadherin was abnormal expression status in ESCC tissues, The positive expression rate of E-cadherin was significantly lower than in adjacent noncancerous tissues, it was negatively correlated with tumor lymph node metastasis and depth of invasion, the detection of E-cadherin has an important clinical significance in forecasting tumor metastasis and evaluating tumor prognosis. (4) The expression rate of Ets-1 had negatively correlation compared with the expression rate of E-cadherin in ESCC, there may be exist antagonism between Ets-1 and E-cadherin in the tumor early onset and infiltration and metastasis of ESCC,combined detection of Ets-1 and E-cadherin can be used as an important referential indexes to predict ESCC early onset, to assess tumor metastasis potential, to evaluate prognosis.
