Total-Flavonoid in Stallera Chamaejsme L was used experimentally to treat and protect the Human Liver Cancer, for observing the antagonistic effect of Total-Flavonoid on tumor and trying to find the way to treat and protect other tumor, based on the antagonistic effect of Total-Flavonoid on tumor. Meanwhile some external trial was on inducing effect on apoptosis of tumoral cell. SMMC-7721 cell was cultured with different concentration of Total-Flavonoid for detected the antagonistic effect on Total-Flavonoid. The relative indexes were detected for expound the antagonistic mechanism of Total-Flavonoid on tumor externally.It is all know that ultrasonic extraction, column chromatography of polyamide are used to extract the plant dry powder in Stellera chamaejasme L.The antitumor activities of the different extracts on liver cancer SMMC-7721 cell line were analyzed using MTT assay. The Result of experiment is show that the total-flavonoid inhibited cell proliferation of liver cancer cell line and its activities are higher than that of others. And showed significant dose-dependent response.MTT assay was used to determine the growth inhibition of SMMC-7721 cells by Total-Flavonoid in Stellera Chamaejsme L. Flow cytometry was applied to detect the change of cell cycle.The expression of caspase-3 was detected by Enzyme-linked immunosorbent assay. It is show that The results of MTT assay indicated that the proliferation of SMMC-7721 cells was significantly inhibited by Total-Flavonoid in Stellera Chamaejsme L in a time-and dose-dependent effect. G2/M phase of cell cycle increased, and the apoptotic peak was cytomeric assay. The percentage of apoptotic SMMC-7721 cells observed by flow change with the concentration of medicine. the expression of caspase-3 in human SMMC-7721cells treated with Total-Flavonoid in Stellera Chamaejsme L obviously increased.The Total-Flavonoid in Stellera Chamaejsme L could induce apoptosis of SMMC-7721 cells. The percentage of apoptotic SMMC-7721 cells is related to the dose and the dose-response relationship is obvious. The growth of SMMC-7721 cells were suppressed by inducing apoptosis, which may be related to the change of the caspase-3 and the cell cycle. It also indicated that emodin probably become a potential anti-tumor drug.
