Objective:To test the manifestation of CyclinD1 protein and STAT3 protein in Oral Squamous Cell Carcimona (OSCC); to clarify the relationship between the different pathological levels of the two in OSCC and their different clinical phases, and the corelation of the two; to explore their function in the occurrence and evolution of OSCC; to assess the value of STAT3 and CyclinD1 in the early diagnosis and after-heal judgement of OSCC, and thereby find new treatment target so as to pave a new way for the genetic treatment of OSCC.Methods:The samples of OSCC are all takes from the pathological department of the First Hospital Attached to Shanxi Medical University from Jan.2005 to Dec. 2009, fifty samples altogether, and the negative control group is composed of 10 normal oral mucous membrances. By employing immunochemical method EnVosion, the project aims to test the manifesting levels of CyclinDl and STAT3 in the pathological sections of the 50 OSCC cases and 10 normal cases. Envision (enhanced labeled-polymer system) PV-6000 Power Vision TM is used to stain and the result is analyzed by using some relevant statistical methods, such as SPSS13.0, X2 test, Fisher and Spearman, P<0.05, and the difference is considered as statistically significant.Results:1.1.In the 50 OSCC samples, STAT3 is maily manifested in cytoplasm, and a little is manifested in karyon. The manifesting level of STAT3 protein (88%) is evidently high above the manifesting level of normal oral mucous membrance (10%) P<0.01, which indicates that STAT3 is an important regulating factor in the occurrence and development of OSCC. The higher tumor differentiation is, the lower STAT3 manifestation is. The high differentiated squamous cell carcinoma has 44.4% manifestion, the middle differentiated squamous cell carcinoma has 73.3%, the low differentiated squamous cell carcinoma 94.1%, P<0.05, and the differences are notable. Meanwhile, the comparison between low differentiation and high differentiation is P<0.01, and the differences are notable, which proves that the positive manifestation of STAT3 and the differentiation levels of oral squamous cell carcinoma are closely related. In the aspect of clinical phases, the positive manifestation of squamous cell carcinoma in clinicalâ…¢phase is 57.1%, in clinicalâ…¡phase is 66.7%, in clinicalâ…¢is 93.3, in cllinicalâ…£phase is 100%, P<0.05, and the differences are notable. Meanwhile, the comparison betweenâ…£andâ… is P<0.01, and the difference is notable, which indicates that the positive manifestation of STAT3 is relevant to the heal of squamous cell carcinoma. The manifesting level of STAT3 in oral squamous cell carcinoma involving lymph node transference is obviously higher than that in the case without lymph node transference involved (P<0.01).2. In the 50 OSCC samples, CyclinD1 is maily manifested in karyon, and a little is manifested in cytoplasm. The manifesting level of oral squamous cell carcinoma is 80%, much higher than that of the normal oral mucous membrance (20%), P<0.01, which indicates that CyclinDl is participating in the occurrence of OSCC. With the increase of the tumor-cell detererioration degree, the CyclinD1 manifestation increases. The high differentiated squamous cell carcinoma has 38.9% manifestion, the middle differentiated squamous cell carcinoma has 66.7%, the low differentiated squamous cell carcinoma 88.2%, P<0.01, and the differences are notable. Meanwhile, the comparison between low differentiation and high differentiation is P<0.05, and the difference is notable, which proves that the positive manifestation of CyclinDl and the differentiation levels of oral squamous cell carcinoma are related. In the aspect of clinical phases, the positive manifestation of squamous cell carcinoma in clinicalâ… phase is 42.8%, in clinicalâ…¡phase is 53.3%, in clinicalâ…¢is 86.7%, in cllinicalâ…£phase is 92.3%, P<0.05, and the differences are notable. Meanwhile, the comparison betweenâ…£andâ… is P<0.05, and the difference is notable, which indicates that it is relevant to the heal of squamous cell carcinoma. The manifesting level of CyclinDl in oral squamous cell carcinoma involving lymph node transference is obviously higher than that in the case without lymph node transference involved (P<0.01). At the same time, in the positive manifestation of OSCC, STAT3 protein and CyclinD1 protein are positively correlative(rs=0.438, P<0.01). Conclusion:1.STAT3 protein and CyclinD1 protein are both participating in the occurrence of OSCC, playing important roles in the occurrence and development of OSCC.2. The manifestation of STAT3 may affect the occurrence and development of OSCC, and is closely related to its differentiation degree, clinical phase and lymph gland transference, so it can be used as a reference for OSCC diagnosis and treatment.3. CyclinDl is closely related to the lymph gland transference, clinical phase, pathological grading of OSCC. The inference of abnormal manifestation of CyclinDl protein may be used as a valuable clinical biological reference for OSCC diagnosis and treatment.4. STAT3 and CyclinDl are positively correlative in the positive manifestation of OSCC, which indicates that STAT3 and CyclinD1 are coordinating or co-regulating themselves in the saturation and transference of tumor and promoting the occurrence and development of tumor together. v...
