BackgroundRecent reports indicated that abnormality of cell cycle is tightly related with the tumorigenesis. The pRb, cyclinDl, CDK4 and p16 proteins, which act in concert to maintain the integrity of the normal cell cycle, are composed of an important cell signal pathway. P16 inhibits CDK4 activity by replacing cyclinDl in the binary CDK-cyclinD1 complex. This inhibition negatively regulates phosphorylation of Rb by the cyclinD1-CDK complex, resulting in inhibiting cell cycle progression. Mutations and deletions of p16, Rb, cyclinDl and CDK4 may disrupt this pathway and result in tumorigenesis.
