Objective To investigate the expression of reversion-inducing cysteine-rich protein with kazal motifs (RECK) and membrane-type1 matrix metalloproteinase (MT1-MMP) in hepatoblastoma and their correlation with metastasis and suppression in the tumor. Methods PV-6000 immunohistochemistry method was used to detect the expression of RECK and MT1-MMP in 35 samples of hepatoblastoma tissue,15 cases of normal liver tissues and 10 cases of benign liver tumor tissues. The SPSS 13.0 software was used in the statistical process. The exact fourfold table x2 test was performed in order to establish the difference among the indicators. The correlation test was applied by the Spearman correlation analysis used for intergroup comparison. Results The expression of the RECK protein in the hepatoblastoma decreased significantly (28.6%), which was significantly lower than the normal liver tissue and liver benign tumor groups, and the comparison between the groups indicated a significant difference (P<0.05).The RECK-positive rate in group hepatoblastoma which has no metastasis was 45%, while that was 6.7%in group hepatoblastoma which has local or distant metastasis. And comparison between the groups indicated a significant difference (P<0.05). Which indicated that the expression of RECK protein reduced with the increase in the depth of tumor invasion and distant metastasis. The expression of the MTl-MMP in hepatoblastoma tissues (54.3%) was significantly higher than that in the normal liver tissues and benign tumor. The positive rate of MT1-MMP expression was 35.0% in group hepatoblastoma which has no metastasis, while that was 80.0% in group hepatoblastoma which has local or distant metastasis. And the comparison between the groups indicated a significant difference (P<0.05), which suggested that MT1-MMP had a high expression in hepatoblastoma tissues, and its expression levels elevated with the increase in the depth of tumor invasion of the tumor and distant metastasis. Conclusions RECK and MT1-MMP may play important roles in the occurrence, development, invasion and metastasis of hepatoblastoma and serve as markers in the estimation of the extent of metastasis and dissemination of the hepatoblastoma.
