Time-resolved fluorescence immunoassay technique is an ultra-sensitive non-radioactive contamination marker analysis technology using lanthanide chelates as markers to measure which has been widely applied in bio-analysis, clinical medicine, food testing.This thesis describes the principle and the development of the status of the time-resolved fluorescence immunoassay and points out the domestic inadequacies of the technique.A solid-liquid phase analysis system with high sensitivity is developed. From the time-resolved fluorescence immunoassay system architecture and performance, many factors effecting the fluorescence signal intensity and SNR are analysed and studied in the following three aspects:1) design of efficient excitation light system; 2) improve filter system to reduce the effect of the background noise,making the inpact of the stray light into a minimal; 3) Selected a more reasonable digital circuits to achieve the sampling integration to improve the signal to noise ratio and the achievement of the adjustment of delay time, the sampling frequency and time.A solid-phase time-resolved fluorescence immunoassay system are designed using Eu3+, Tb3+ as markers, the system has advantages of a high sensitivity and good stability.The sensitivity is testing to be 10-13mol/L,in the range of 10-12-10-9mol/L,the fluorescence intensity maintain linear,and the stability CV≤2.5%.This is a quantitative micro amount detection of clinical testing.
