Objective Through comprehensive study about influence factors on the quality of medicinal Chrysanthemum such as different picking periods, different flowering,different processings, different storage times and different areas and so on,we can provide the scientific basis for the rational harvest processing and quality control of Chrysanthemum.Methods (1) Use volatile oil extraction device to compare different flowerings,different processings and storage times of content of volatile oil in Zhejiang Medicinal Chrysanthemum, and GC-MS technology to analysis components of the essential oil. (2) Use rutin and glucose as the control, the establish separately of total flavonoids and polysaccharides in UV spectrophotometric determination, compare content of total flavonoids and polysaccharides of different flowering, different processings, storage times and different areas in Zhejiang Medicinal Chrysanthemum. (3) Use luteolin, chlorogenic acid as the control, establish HPLC determination method of luteolin and chlorogenic acid separately and compare the different flower, different processing, different storage time and different areas of the Osmanthus Chrysanthemum grass elements and chlorogenic acid. (4) Use agar diffusion method in the KB method (Kirby-Bauer disk diffusion method) for Chrysanthemum in vitro antibacterial test.Results By GC-MS technical analysis,74 peaks were separated from essential oil, identify 25 compounds, which are 33.78% of the total composition; volatile oil has obvious changes in Chrysanthemum of different flowerings and different storage times.Different picking periods,different processing and different areas obviously affect the content of flavonoids in Chrysanthemum;different flowering and different storing time have no effect on the content of flavonoids.There are obvious changes in content of polysaccharides in Chrysanthemum of different picking times,different flowering and different areas. Different picking periods,different flowers has obviously influence the content of chlorogenic acid;different processing,different storing times,different areas has no obviously effect on the content of chlorogenic acid. There are obvious changes in content of luteolin of different picking flowering,different flowering and different processing. Essential oil on a variety of common infectious bacteria have antibacterial activity significantly.Conclusion Chrysanthemum was established in this study of total flavonoids and polysaccharides in quantitative UV spectrophotometric method and luteolin and chlorogenic acid in the HPLC quantitative method, the method's stablity, precision, reproducibility is good,which can test the quality of medicines fast, accurately and simplely.It can offer experimental evidence for quality control of medicines. Chrysanthemum active ingredient is obviously iffluenced by picking, tubular flower open, processing methods, storage time and original areas. The first flower, Chrysanthemum fixing method for processing have good quality. The study provide a scientific basis for Chrysanthemum harvest processing and reasonable original areas choice.
