Objective: The aim of this study was to investigate the changes of recurrent oral ulcers and interleukin-6 levels with vitamin B12(one of simple, inexpensive, low-risk drugs)by animal level, so as to provide a new theoretical basis to the cause and clinical treatment of recurrent oral ulcers.The present study was undertaken to evaluate the treatment effectiveness and investigate the mechanism of vitamin B12 in the oral ulceration by observing the changes of oral ulcers(ulcer number, duration, interval) of experimental rats that treated with vitamin B12 and testing IL-6 levels in peripheral blood serum.Methods: Rats models of recurrent oral ulceration with immunology were established. Add Freund's complete adjuvant(FCA) to galacta of health variant rat's oral mucosa as the antigen, and injected it to two sides of spinal column, 0.1ml per side, one time two weeks, oral ulcers appear one week after the third injected. While established the control group(injected the normal saline and FCA), cut the oral mucosa of normal and ulcer rats, and make Hematoxylin-eosin(HE) staining pathology examination; These ulcer rats (male and female) were randomly divided into ulcer group, vitamin B12 group and the levamisole group. Intragastric administration was before taking food with the dose of 20μg/Kg of Vitamin B12 group and 20mg/kg of levamisole group , and other groups are normal diet. Drawing blood from the eyes of the rats after 30 days intragastric administration and centrifugating , and detecting serum IL-6 levels by enzyme-linked immunosorbent assay. Compared by least significant difference t test: ulcer group and the normal group, vitamin B12 group and the ulcer group, the levamisole group and the ulcer group.Results:1.Ulcers were formed by observed and histopathology examination. Histologically, infiltration of intraepithelial lymphocytes, dissolved ulceration, a large number of inflammatory cell infiltration in lamina propria, capillary congestion expansion, gland hyperplasia, the ulcers were formed;2.IL-6 levels were measured by ELISA, calculated the concentration by the standard curve, the outcome of statistical analysis were: The IL-6 levels of the ulcer group were significantly higher than normal, the difference was statistical(p<0.05); The IL-6 levels of vitamin B12 group and levamisole group were decreased, close to the normal group, the difference was statistical(p <0.05).Conclusion:1.The serum IL-6 levels of recurrent oral ulcers group were increased compared with normal group, suggesting that cellular immunity may be involved in the pathogenesis of recurrent oral ulcers;2.Vitamin B12 may regulate rats'immune system by reducing IL-6, to promote healing and reduce recurrence.
