Objective The technology of ultra-fine powder is developed recently and shows many advantages in Chinese medicine preparation,herbal slice,extracts. In order to study the application of this technology in traditional Chinese medicine,we should establish the modern quality standards for the ultra-fine powder, especially about the reasonable particle size control standards and quality evaluation system for each sample.The research was carried out on the basis of traditional quality evaluation system of TCM and various modern analytical approach and evaluation technology were adapted to explore the multi-component, multi-target effects mechanism that Flos Lonicerae produced.Through the research,we can see the effective particle size range and the most effective active substance concentrated area.Method Traditional heat-clearing and detoxifying drug—Flos Lonicerae powder was used as the research object,and we have done the research by following methods:1. In our study,the ultra-fine time,temperature,water content,medium fill rate index were selected using D50 value measured,the image and laser methods were used to identify the micro-morphous and the grain size of ultra-fine powder; 2.The best efficiency powder of two particle size powder can be compared obviously by the chemical composition dissolution studies,the GC-MS analysis for essential oil samples and vitro antibacterial activity assay; 3.Common model fingerprint were established by HPLC-DAD and GC,combined with multi-target ingredients quantitative analysis; 4. Six different polarity extracts were obtained using the solvent system and relative retention time and relative peak area were standardized,the most valid extract was chosen by the Correlation analysis about Ri-αi data and antibacterial activity results.Results 1. The optimization ultra-fine condition of Flos Lonicerae was deter-mined:crushing time was 7min,temperature was-10~10℃,water content was 5~7%,medium filling rate was 60~70%;the optimal particle size distribution was D50≤16μm,The normal microscopic characteristics could not be seen from the ultra-fine powder; 2.The study on the dissolution in vitro of chemical components indicates that the quanitity of extract and rate of dissolution speed of the ultra-fine powder were enhanced;Ultra-fine powder can be regarded as the comparative advantage powder by the GC-MS and the antibacterial activity analysis; 3. The GC fingerprint was established after the analysis of 8 batches essential oils from Flos Lonicerae to compare the similarity among samples from different origins,36 peaks were determined as common fingerprint peaks;Methanol extracts of Flos Lonicerae from 10 different origins were analyzed with HPLC,and 27 common peaks were confirmed by using the gradient elution. The data was analyzed by "Computer Aided Similarity Evaluation software" recommended by Chinese Pharma-copoeia Commission.The similarity of fingerprints and clustering of 10 different samples were mathched; 4. Relative peak area fingerprint characteristics of different extracts and the Correlation analysis about Ri-αi data and antibacterial activity results were established. Conclusion During the ultro-fine process of Flos Lonicerea,the appearance, microscopic characteristics,chemical components content and the dissolution behavior were changed more or less,however,the total dissolution and bioavaila-bility were increased in ultro-fine powder. The quality assessment of Flos Lonicerea was systematic established and the most effective active substance concentrated area was explored. Bioactive-fingerprint of relative peak area for Flos Lonicerea powder can be established by the Ethyl acetate extracted part.The results validated the superiority of ultro-fine powder and pave the way for optimizing development and application of Flos Lonicerea.
