| Decoction of traditional Chinese medicine is one of the main ways of clinical application of traditional Chinese medicine.With thousands of years of application experience and theoretical knowledge support,decoction can be used easily with symptoms.However,due to the rough and different drug forms,uneven quality,difficult to control the quality of the problem,and the way of decoction and taking cumbersome,limited the promotion of traditional Chinese medicine in the modernization of traditional Chinese medicine.The ultra-fine powder retain the material basis and properties of the original traditional decoction pieces;improve the uniformity;further accelerate the dissolution rate of the ingredients,improve the utilization rate of the ingredients,and reduce the dosage.However,due to the change in the physical properties of the quality of the pieces,such as the angle of repose and the hygroscopicity,from the traditional decoction pieces to the ultra-fine powder,it is necessary to take measures to improve the quality of the products.Dendrobium officinale ultra-fine granular is a full-component granule of 30-100 mesh pieces prepared by using Dendrobium officinale ultra-fine powder without adding auxiliary materials.It not only retains the advantages of ultra-fine powder,but also overcomes the problem of easy oxidation,moisture absorption and agglomeration of ultra-fine powder.In this paper,Dendrobium officinale was selected as the research object,and the preparation process and physical chemical characteristics of Dendrobium officinale powder were studied.The comparative analysis between Dendrobium officinale common powder,ultra-fine powder and ultra-fine granular powder was studied.The quality evaluation study provides a certain experimental basis for the development of new formulations of Dendrobium officinale.(1)The preparation process of Dendrobium officinale was based on D90 as the test index,combined with the determination of polysaccharide content,using single factor and superfine pulverization process.Preferably,the ultrafine powder pulverization process has a moisture content of 7.5%,a material ratio of 3:1,a pulverization temperature of-4℃,and a pulverization time of 5-7 h.Characterization and evaluation of different particle size ultra-fine pieces of Dendrobium officinale.From the physical index such as particle size distribution,specific surface area,angle of repose and et al.The results showed that the Dendrobium officinale was ultrafine crushed,its particle size decreased,specific surface area,loose density and hygroscopicity increased,sliding Angle,rate of flow,water holding capacity and swelling force decreased,and oxidation,hygroscopicity and agglomeration were more likely to occur.(2)To investigate the effects of different alcohol concentration and drying temperature on the preparation process of Dendrobium officinale,and to screen out the excellent production process of ultra-fine granular powder.A comparative study was conducted on common powder,ultra-fine powder and ultra-fine granular powder.Comparisons were made from indicators such as traits,identification,inspection,particle size distribution,hygroscopicity,and dissolution in vitro.The results showed that the common powder of Dendrobium officinale changed significantly after ultrafine pulverization;with the decrease of particle size,the hygroscopicity increased,and the t common powder<ultra fine granular powder<ultra fine powder,after granulation into granules,Its hygroscopicity was reduced within a certain range;as the particle size decreases,the dissolution of active ingredients increases,and the ultra fine granular powder>ultra fine powder>common powder,showing that the ultra-fine powder can be effectively processed by granulation into pieces.The elution amount of the component promotes the dissolution rate,which indicated that the gelatinization and aggregation of the powder are reduced after granulation,and the dissolution of the active component is more favorable.(3)The fingerprint of Dendrobium officinale ultra fine granular powder was established by HPLC.The results of methodological investigation showed that the method was stable and reliable,and could be used to control the quality of Dendrobium officinale.Through fingerprint analysis of 10 batches of different Dendrobium officinale materials,22 common peaks were selected,and similarity evaluation was carried out by using the similarity evaluation system of traditional Chinese medicine chromatogram fingerprint.The results showed that the overall quality of Dendrobium officinale was significantly different among different batches,indicating that the content of effective components of Dendrobium officinale was significantly different among different batches.(4)This paper describes the appearance of Dendrobium officinale ultra fine granular powder,identifies the microstructure of Dendrobium officinale ultra fine granular powder,determines the content of water,extract and mannose in Dendrobium officinale ultra fine granular powder,and determines the particle size distribution of Dendrobium officinale ultra fine granular powder.The quality evaluation system of Dendrobium officinale was established. |
