| Objective To explore the effects of Qin Xiang diarrhea enteric-coated tablets on the diarrhea-irritable bowel syndrome of plasma VIP, NPY and mucosal 5-HT.Methods 44 SD rats of clean grade were evenly divided by male and female, body mass 220±20g. After fed 7 days adapting to the environment, they were stochasticly divided into four groups, such as blank group, model group, Qin Xiang diarrhea enteric-coated tablets group and loperamide hydrochloride group. The remaining four SD rats were used to observe the success of the model.The blank group was given normal feed and was not involved in modeling, the other groups were applied with Lio France spleen diarrhea and chronic mild stress increases keep France isolated in order to stablish the diarrhea-irritable bowel syndrome in rats. After models were successful, administrational groups were given Qin Xiang diarrhea enteric-coated tablets and control groups were given Loperamide hydrochloride liquid, the blank group and model group were given normal saline, administered for 7 days. The animals were killed and taken blood and colon, ileocecal organization, observed the change of plasma VIP, NPY, and intestinal tissue 5-HT in rats.Results 1. Qin Xiang diarrhea enteric-coated tablets impacted on the D-IBS rats's general states:The rats after modeling became apathetic,fatigue, unresponsive, stool thin pond, pond leaks, perianal contamination, declining in food's intake and weight, slow growth, issued dry dull fur, vertical hair, IBS model group became irritability, sugar intake decreased significantly. Qin Xiang treatment group and the positive drug control group were fed with the appropriate medications, the symptoms improved significantly.2. Qin Xiang diarrhea enteric-coated tablets effected on the D-IBS plasma VIP,NPY:the plasma VIP levels of model group were significantly higher than the control group, the plasma NPY levels were significantly lower than the control group, the difference was statistically significant (P<0.05); by drug intervention, the plasma VIP levels of Qin Xiang enteric-coated tablet group and loperamide hydrochloride group were significantly decreased comparad with the model group, NPY levels were significantly increased, the difference was statistically significant (P<0.05); The comparison plasma VIP, NPY level were not significantly different between Qin Xiang enteric-coated tablet group and loperamide hydrochloride group (P>0.05). Qin Xiang enteric-coated tablets can improve the symptom of model group, the effects is as the same as the Loperamide hydrochloride liquid. 3. Qin Xiang diarrhea enteric-coated tablets effected on the D-IBS intestinal tissue 5-HT in rats:Comparing with the normal group, the 5-HT positive cells of model group were significantly increased, the immunohistochemical staining were significantly enhanced, the difference was statistically significant (P<0.05);by drug's intervention, the 5-HT's positive cells and staining intensity of Qin Xiang enteric-coated table group and hydrochloride loperamide group were significantly reduced comparing with the model group (P<0.05), no significant difference was compared with blank group (P>0.05).Conclusion 1. With the way of Lio France spleen diarrhea and chronic mild stress increases keep France isolated,we created the diarrhea-irritable bowel syndrome rat model successfully.2. The plasma NPY,VIP of D-IBS rat model were changed, the distribution of intestine 5-HT showed abnormal, suggesting that these brain-gut peptide were relevance to the D-IBS.3. Qin Xiang diarrhea enteric-coated tablets could regulate the release of VIP,NPY,5-HT, reduce the gastrointestinal motility, reduce the intestinal inflammation, reduce intestinal sensitivity to improve diarrhea. It could provide a reliable theoretical and experimental evidence for treatment of the irritable bowel syndrome. However, the specific pathway should be further explored. 4. Qin Xiang diarrhea enteric-coated tablets owe the effect of heat dampness, Shibuya intestinal diarrhea, Qi analgesic efficacy,It is the valid prescriptions.which is used to cure the diarrhea-irritable bowel syndrome...
