Experimental Study On Er Huang Yu Yang Powder In A Rat Modle Of Infected Cutaneous Ulcer Healing

Objective:This present study is to investigate whether Er Huang Yu Yang Powder can promote wound healing and its mechanism and to find a new remedy for treating the infected skin ulcer and provide the pharmacological basis of the new remdy.Methods:1. Skin toxicity and skin sensitization test; 2. Study on the role of promoting heal, Rats after general anesthesia back hair removal zone steam iron 40 seconds, so that cause ulcer model, Burn-zone cooling 15 minutes before injection of E. coli to modeling, to become infected ulcer model.After five days of modeling,the wound surface crusts removed, and put them into the infected ulcer group and the non-infectious ulcer group, they were given treatment by Er Huang Yu Yang Powder, at the same time with Kang Fu Xin treatment group and saline treatment group as the control.The main observation indicators are Local dynamic observation of wound and Histomorphology, When wound healing using immunohistochemical methods and Image-Pro-Plus image analysis system for epidermal growth factor (EGF), transforming growth factor (TGF-β1), tumor necrosis factor (TNF-α), Interleukin-8 (IL-8) quantitative analysis of protein expression.Result:1. Er Huang Yu Yang Powder was no stimulation of skin in rabbits, no sensitization of the guinea pig skin; 2. The healing time of Er Huang Yu Yang Powder treatment group compared with Kang Fu Xin treatment group is shorter in the infected ulcers.3. The healing time of Er Huang Yu Yang Powder treatment group Kang Fu Xin treatment group is equivalent with Kang Fu Xin treatment group in the non-infectious ulcers.4.The IL-8 of Er Huang Yu Yang Powder treatment group are below compare to Kang Fu Xin treatment group and saline treatment group in the infected ulcers,and EGF Significantly higher than that of Kang Fu Xin treatment group and saline treatment group in the infected ulcers.5. The IL-8 and TNF-αof Er Huang Yu Yang Powder treatment group are below compare to Kang Fu Xin treatment group and saline treatment group in the simple ulcers,and EGF Significantly higher than that of Kang Fu Xin treatment group and saline treatment group in the simple ulcers.Conclution:1.Er Huang Yu Yang Powder is no Toxicity to Rat skin;2. Er Huang Yu Yang Powder is no Sensitization to Rat skin;3. Er Huang Yu Yang Powder has significant anti-inflammatory and promote healing of infected skin ulcer, The anti-inflammatory of Er Huang Yu Yang Powder is better than Kang Fu Xin and is same in the healing of Kang Fu Xin.4.The mechanism of its efficacy may be mainly related to reduced TNF-αand IL-8 expressions and significantly increased EGF expressions, and on the expression of TGF-β1 had no effect. Er Huang Yu Yang Powder scattered down the expression of IL-8 increase the expression of EGF is superior to Kang Fu Xin.
