Research On The Chemical Composition And Quality Standard Of Platycodons Radix And Its Herbal Prepared For Decoction

Platycodon grandiflorum(Jacq.)A.DC.is the dry root of Radix Platycodonis.It has a saline and acrid flavor,mild attribute.It is a medicine most in use of relieving cough and reducing sputum.It is also known as Bell flower,Monk hat,Baiyao,Talinum crassifolium,Doraji and Kikyo and so on.Its officinal root has a saline and acrid flavor,mild attribute,It has the function of ventilating lung,relieving sore throat,eliminating phlegm and draining pus. Platycodon grandiflorum(Jacq.)A.DC.is renascent herbs,growing in desolate mountains, underbrush and shrub.The main producing region includes Anhui, Henan, Hubei,Liaoning,Jilin,Hebei,and Nei Menggu province.North-east and North China produce most and the production is called"north Platycodon",the East China produces the best and is called"South Platycodon".This paper has done extraction and separation of the chemical ingredients of platycodon.In the experiment the author marinated Platycodon grandiflorum(Jacq.)A.DC.in 70% ethanol.The soak solution was condensed into drying powder,manufactured into drying powder of platycodon extractive.Then it was dissolved in the water and used the D101 macroporous resin,then orderly did the elution by water,20% ethanol,40% ethanol,and 95% ethanol,then still collected the 40% ethanol part,condensed in the rotatory evaporator and then makes it frozen drying powder by refrigeration compressor.Using silica gel column chromatography,the author did gradient elution with chloroform-methanol-water system,and checked the regenerant by TLC and high performance liquid,then coalesced fourteen parts and then purified different part with prep-HPLC,acetonitrile-water being the mobile phase,the eluant was frozen by refrigeration compressor.The author got chemical compoundâ… 51.83mg,chemical compoundâ…¡9.0mg,chemical compoundâ…¢10.16mg,chemical compoundâ…£22.70mg.By the physicochemical property of the chemical compound and the bop data of NMR,the author examined the structure of the chemical compound,respectively, Platycodin D, Platycoside G3,Lobetyolin. Platycodin is the main effective active ingredient of platycodon grandiflorum(Jacq.)A.DC.According to Chinese Pharmacopoeia 2005,the mensuration of total saponin content of platycodon grandiflorum(Jacq.) A.DC. is gravimetric method,and formulates that the total saponin content cannot be less than 6%.This mensuation is complex,low-specificity,poor reproduction quality and the experiment result has big inaccuracy and it is hard to examine the quality of the medical herbs and herbal pieces prepared for decoction.The thesis controlled the quality of platycodon grandiflorum(Jacq.)A.DC.by TLC and ELSD detector with treating the platycodin D extractive from laboratory as the reference substance.Moreover,Chinese Pharmacopoeia 2005 does not give formulation of the moisture content,total percent of ash,acid-insoluble ash and extractive.The thesis did above mentioned mensuration of thirteen batch of platycodon grandiflorum(Jacq.)A.DC.herb material and herbal pieces prepared for decoction by the method captioned by pharmacopoeia and fixed the test standard of moisture content,total percent of ash,acid-insoluble ash and extractive of platycodon grandiflorum(Jacq.)A.DC. herb material and herbal pieces prepared for decoction.The thesis provides much effective data of the production of platycodon grandiflorum(Jacq.)A.DC. herb material and herbal pieces prepared for decoction to Chinese Pharmacopoeia 2010.Now the standard is recorded in Chinese Pharmacopoeia 2010.
