The roots of Umbelliferae Heracleum candicans Wall,ex DC was used as Zhuga for medicinal by Tibetan.The plant morphology of Heracleum candicans Wall.Ex DC was consistent with the description in and other books.Zhuga had the effects of insecticidal,hemostasis,curing carbuncle and leprosy.By the research group in Sichuan Province,Ganzi,Aba Prefecture survey found that Heraclei candicantis Radix had good curative effect,was rich in resources.Therefore,it is of great significance to further research and development of Heraclei candicantis Radix.So far,the domestic and foreign research of Heraclei candicantis Radix was little,no quality standard research.At the time,The same survey found that the dried root of a originates from Umbelliferae Angelica genus Heracleum souliei de Boiss.were often mixed with Zhuga medicinal,medication caused confusion,the safety of clinical medication effects.This study was required by the standard basis of Sichuan province food and Drug Inspection Institute of scientific research "to develop Sichuan Tibetan medicine".While Completed of Heraclei candicantis Radix quality standards,including the different origin of Heracleum candicans medicinal properties,identification,inspection,extract,content determination and pharmacology were studied to provide scientific basis for the quality evaluation of Heraclei candicantis Radix.Comparative Study was carried on Heraclei candicantis Radix and Heraclei Souliei Radix from the original plant morphology,properties,identification,determination and pharmacological.The specific research contents and results are as follows:The traits of Heraclei candicantis Radix and Heraclei souliei Radix were identified.The root color,shape,section,smell were the main characteristics.Comparison of Heraclei candicantis Radix and Heraclei Souliei Radix,the major difference was the root of the color and smell.The experimental study of identification at Heraclei candicantis Radix.According to the 2010 edition of Chinese Pharmacopoeia established the TLC identification method of appendix HPLC,fingerprint identification method.TLC identification of taking imperatorin as control sample,the sample volume 4 L,respectively in the same silica gel G plate and GF254 plate,the agent of petroleum ether(60~90℃)-ethyl acetate(7:3),the 10 batch of Heraclei candicantis Radix in the corresponding position of silica gel G plate and GF254 plate showed the same color spots.HPLC fingerprint for the identification of the bergapten,imperatorin as reference,using Diamonil C18 column(250*4.6 mm,5 μm),the mobile phase was A for water B methanol,gradient elution:35%B maintains 5 min,35%-95%B 55 min,95%B 5 min,the column temperature was 30℃,the flow rate was 1 mL/min,the detection wavelength was 302 nm,at time 65 min,sample volume 20μL,Results the common mode of HPLC fingerprints of Heraclei candicantis Radix.a total of 11 peaks;fingerprint and fingerprintsimilarity was greater than 0.9,good similarity.The comparative study between Heraclei candicantis Radix and Heraclei souliei Radix by use of the method of Heracleum candicans TLC and HPLC fingerprint for the identification,the results of the two difference was different,could be better identified.The experimental study of inspection item at Heraclei candicantis Radix.According to the method in the appendix of 2010 edition of Chinese Pharmacopoeia,on 10 different batches of Heraclei candicantis Radix samples of water,total ash,acid insoluble ash content inspection.According to the experimental results,the proposed limits of Heraclei candicantis Radix sample:water should not be more than 12%;the total ash not more than 8%;the acid insoluble ash shall not exceed 2%.The experimental ’study of extractive content at Heraclei candicantis Radix,According to the method in the appendix of 2010 edition of Chinese Pharmacopoeia,Determination of 10 different batches of Heraclei candicantis Radix samples were extract with 30%ethanol extraction method.Preparing extract content of not less than 28%.The experimental study of determination items at Heraclei candicantis Radix.established the determination method of total coumarin and two active ingredients.With UV-VIS absorbance was measured at 290 nm,the calculated on dry goods,including the total content of coumarin to psoralen meter range 2.29-4.76%(average 3.40%).The total coumarin content of not less than 2.5%formulation.established a total coumarin content method of Heraclei souliei Radix,calculated on dry goods,including the total content of coumarin to psoralen meter range from 0.62 to 2.53%(average 1.53%).Results:the big difference between the content.By the method of HPLC like HPLC fingerprint with the conditions established method to measure two active ingredients content of Heraclei candicantis Radix which included bergapten and imperatorin were 0.12%,0.71%.While established a method for the determination of the bergapten and psoralen of Heraclei souliei Radix.the result content psoralen,bergapten were 0.08%,0.12%.Comparison of Heraclei candicantis Radix and Heraclei souliei Radix,contain similar content of psoralen.But Heraclei candicantis Radix did not contain psoralen;Heraclei souliei Radix contained a trace amount of imperatorin.The pharmacology experimental study of acute toxicity,bleeding,anti-inflammatory and analgesic about Heraclei souliei Radix and Heraclei candicantis Radix.The results show the fed MTD of Heraclei candicantis Radix and Heraclei souliei Radix were 17.8 g/kg and 16.8 g/kg.the maximum tolerated dose was approximately 118 times the amount of adult and 112 times.the high,medium and low dose group of Heraclei candicantis Radix and Heraclei Souliei Radix could reduce dimeth-ylbenzene swollen auricles of mice in trend,the high,medium and low dose group of Heraclei Souliei Radix could significantly reduce the mice writhing,there was not significant differences in the high,medium and low dose group of Heraclei candicantis Radix but trend.the high and medium dose group of Heraclei candicantis Radix could significantly make the clotting time short.The original plant comparative study about Heraclei candicantis Radix and Heraclei souliei Radix found that the difference mainly in the original plant fruits and leaves.The chemical components pre-experiment about Heraclei candicantis Radix and Heraclei souliei Radix.The results show that both contain ingredients coumarin,volatile oil,polysaccharides,organic acids.The volatile oil GC-MS analysis about Heraclei candicantis Radix and Heraclei souliei Radix.The results show that the two volatile oils were not the same.Through the quality standard study of Zhuga(Heraclei candicantis Radix),the initial establishment of Heraclei candicantis Radix draft standard,comparative study of Heraclei candicantis Radix and Heraclei souliei Radix,further development of both resources and laid the theoretical foundation of traditional Chinese medicine to increase resource utilization and resource guide has practical significance for sustainable development.
