The extraction and separation process of protopanaxatriol-type a-glycosides (ATG) in Sanqi was investigated. The methanol extract of Sanqi was separated over D-101 macroporous resin followed by MCI gel to yield the fraction enriched ATG. The obtained a-glycoside fraction contained 8 PPT-type glycosides (ATG-1-8). By comparing retention time with authentic compounds,5 of the 8 glycosides were characterized, among which ATG-2,5,7,8 were a-glycosides, and ATG-4 was P-glycoside. Furthermore,3 glycosides, ATG-1,4, and 6 were isolated form ATG fraction. The structures of ATG-1 and ATG-6 were preliminarily deduced based on their spectroscopic data.The effect of pH value on the kinetic process of water-extraction of Danshen was also be investigated. After extracted by water with different pH value, the contents of danshensu, protocatechuic aldehyde and salvianolic acid B in Danshen extract were determined using high performance liquid chromatography. Through the comprehensive consideration on extraction and decomposition of salvianolic acid B, a water-extraction kinetic model was established. Furthermore, the kinetic parameters with various pH values were calculated. With the pH value elevating, salvianolic acid B content decreased while danshensu and protocatechuic aldehyde contents increased. And the decomposition rate constant of the salvianolic acid B increased, while the extraction rate constant decreased. As a result, the pH value of extraction solvent is important for the stabilization of the ratios of danshensu, protocatechuic aldehyde and salvianolic acid B in Danshen products.
