Objective: To detect the expression of Ki-67,MMP2 and the relationship of them in invasive pituitary adenomas, and to investigate whether there exist correlations among Ki-67,MMP2 and the invasiveness of invasive pituitary adenomas, to explore the correlation between the invasiveness and texture of pituitary adenomas(PA). which may help us diagnose, treat pituitary adenomas and predict the patients,prognosis.Methods: 76 cases of PA being verified surgically and pathologically were collected at our hospital from January 2008 to January 2010,then were divided into two different groups based on different criteria: invasive group(32 cases) and non-invasive group(44 cases) according to their MRI finding; hard group(22 cases) and soft group (54 cases) in texture according to surgical finding. HE staining was use to observe the organization,structure and arrangement of pituitary adenoma cells. Immunohistochemical staining (Envision System method) was used to detect the expression of Ki-67 and MMP2 in these samples of PA. statistically analysing the correlation between the invasiveness and the expressions of Ki-67 and MMP2, and the correlation between the invasiveness and the texture of PA.Results:①Pathological features of HE staining between invasive and non-invasive pituitary adenomas are not easy to distinguish under the light microscope, which shows the cell heteromorphism, large nuclear, nucleoli evident, the significant proliferation of mesenchymal cell and the hyperplasia of the capillary.②The expression of Ki-67 and MMP2 in invasive group is significantly higher than the one of non-invasive group (P<0.01). Ki-67 is positively correlated with MMP2 (r=0.476, P<0.01).There is no correlation being found between the invasiveness and the texture of PA(χ2=3.665, p=0.056)。Conclusion: Ki-67 and MMP2 can be used as molecular markers to evaluate the invasiveness of PA .Detecting Ki-67 and MMP2 can help us build individual therapy and judge prognosis .The texture of PA can not be determined simply by its invasiveness presurgically.
