Objectives:Research about the effect of Herceptin combined with oxaliplatin to invasion of NCI-N87 gastric cancer cells which is positive to HER-2/neu.Methods:1. Transwell invasion chamber assay test cell migration and invasion of NCI-N87 cells after drug effects.2.Using MTT colorimetry assay adhesion ability of cell.Results:1. The results can be seen from the different concentrations of Herceptin alone, the NCI-N87 gastric cancer cell invasion was not found significantly inhibited (P> 0.05)。The effect of different concentrations of oxaliplatin on the NCI-N87 cells 48h after the invasion of reconstruction basement membrane inhibition rates were 9.22%,24.12%,44.29% and 62.81%.Compared with the control group, the difference was statistically significant (P<0.05) and correlation analysis showed that the dose-response relationship (P<0.05).With a final concentration of Herceptin and oxaliplatin 100ug/ml (2,4,8,16ug/ml) for the treatment of gastric cancer cells 48h, inhibition rates of reconstruction basement membrane invasion were 35.10%,49.04%,62.77%,78.46%.Compared with oxaliplatin alone, the difference was significant (P<0.05).2. The results can be seen from the use of different concentrations of Herceptin alone, the NCI-N87 gastric cancer cell migration inhibitory effect was not found significant (P> 0.05).The effect of different concentrations of oxaliplatin on the NCI-N87 cells after 48h of migration inhibitory rates were 12.71%,26.78%,37.25%and 58.71%. Compared with the control group, the difference was statistically significant (P<0.05) and correlation analysis showed that the dose-response relationship (P<0.05).With a final concentration of Herceptin and oxaliplatin 100ug/ml (2,4,8,16 ug/ml) for the treatment of gastric cancer cell 48h,the inhibition rates of migration were 30.65%,44.80%,52.11%and 65.13%.Compared with oxaliplatin alone, the difference was significant (P<0.05).3.The results can be seen from the use of different concentrations of Herceptin alone. the significantly inhibited of NCI-N87 gastric cancer cell adhesion to the Matrigel were not found (P> 0.05).The effect of different concentrations of oxaliplatin on the NCI-N87 cells after 48h of adhesion inhibitory rates were 10.99%.25.27%,43.96% and 64.35%. Compared with the control group, the difference was statistically significant (P<0.05) and correlation analysis showed that the dose-response relationship (P<0.05).With a final concentration of Herceptin and oxaliplatin 100ug/ml (2,4,8,16 ug/ml) for the treatment of gastric cancer cell 48h,the inhibition rates of adhesion were 17.58%,40.66%,62.64% and 76.92%.Compared with oxaliplatin alone, the difference was significant (P<0.05).Conclusion:Although Herceptin cannot apparently inhibit invasive metastasis of gastric cancer cells, it can enhance inhibition of oxaliplatin to gastric cancer cell which is positive to her2.
