Objective:To study the inhibitory effects that herceptin alone and combined 5-FU or oxaliplatin affect on gastric cell line,and provide a certain theoretical basis for herceptin combined chemotherapeutics on clinical therapy.Methods:Select her2 positive gastric cell line(NCI-N87) and her2 negative gastric cell line(MGC803) as the object of study.MTT assay was carried examine the inhibition ratio of herceptin 5-FU oxaliplatin,alone and herceptin combined with 5-FU or oxaliplatin,with various concentrations on gastric cell line for 48 hours.Results:The results of two separate application Herceptin gastric cancer cells were no apparent inhibition. Two chemotherapy drugs 5-FU and oxaliplatin into two kinds of stomach cancer cells had an significant inhibitory effect, and with the increase of drug concentration, with inhibition enhance.To NCI-N87 lines,the maximum inhibition rate of 5-FU was 73.7% and oxaliplatin was 50.2%.To MGC803 lines,the maximum inhibition rate of 5-FU was 54.4% and oxaliplatin was 47.9%.Herceptin combination chemotherapy drugs effect on NCI-N87 lines enhance the chemotherapy drugs inhibition.The combined chemotherapy drugs inhibition rate increases, greater than simple addition of drugs,Maximum inhibition rate 5-FU98.7%(p< 0.05),oxaliplatin 90.6%(p<0.01). Herceptin can obviously increase that to her2 positive gastric cancer chemotherapy drug inhibition of cells. And in the role of Herceptin combination chemotherapy drugs on MGC803 lines and used alone chemotherapy drugs role was not statistically significant difference (p>0.05).Conclusion:5-FU or oxaliplatin has a increasing concentration with inhibition enhanced cell toxicity on Her2 positive (NCI-N87) and negative (MGC803) of stomach cancer cell. There was no significant inhibiting effect on MGC803 and NCI-N87 lines when only used Herceptin,meanwhile, combination chemotherapy drug 5-FU or oxaliplatin also can not increase her2 negative lines MGC803 inhibition, but for Her2 positive NCI-N87 lines, the combination chemotherapy can obviously increase the chemotherapy drugs toxic effects of the cancer cells.
