The Experimental Study Of Herceptin Combined With Oxaliplatin On Gastric Cancer In Vitro

Objectives:1. To study the influence of Her-2/neu monoclonal antibody on the proliferation of Her-2/neu positive and Her-2/neu negative gastric cancer cell lines in vitro, and to evaluate the application value of the therapy of gastric cancer.2. To evaluate the synergistic effect of Herceptin combined chemotherapy drug Oxaliplatin to gastric cancer cells lines so as to ascertain the optimal therapy schedule and to provide experimental evidence for clinical treatment.Methods:1. Detecting the expression of Her-2/neu on the human gastric cancer cell lines NCI-N87, SGC-7901 by RT-PCR method.2. Detecting the inhibitory effects of Herceptin,chemotherapy drug alone or in combination on gastric cancer cells by CCK-8 assay.3. Flow cytometry was used to determine the effect of Herceptin and OXA alone or in combination on cell cycle and on the apoptosis of human gastric cancer cells4. TUNEL was to detect the apoptosis ratio of cells with the different experimental groups.5. Western-blot method was used to detect the influence of Herceptin and OXA alone or in combination to the protein expression of AKT,pAKT,NF-κB on two kinds of gastric cancer cells.Results:1. Human gastric cancer cell line NCI-N87 showed positivity of Her-2/neu expression, and SGC-7901 showed negativity.2. No growth inhibition was observed both NCI-N87 and SGC-7901 cells,when Herceptin was used alone.3. Herceptin augments the cytotoxicity of Oxaliplatin when given after chemotherapeutic agents in NCI-N87,but not in SGC-7901 cells.4. Herceptin alone or combined with Oxaliplatin can inhibit the expression of pAKT in N87,and Herceptin in combination can improve the expression of NF-κB protein and all drugs alone or combined showed no influence on the expression of AKT. Conclusion:Herceptin alone had no growth inhibitory effects on gastric cancer cells in vitro,but can increase cytotoxicity and exist the optimal schedule by following Oxaliplatin.
