Preeclampsia is a specific disease during pregnancy, is characterized by hypertension proteinuria and other systemic disorders,is one of the main reasons of leading to maternal and perinatal disease and mortality, seriously threatens maternal and child health.So far,there are a variety of theories about the causes and pathogenesis of preeclampsia,but these theories are not fully clarified. Recently, most of the research at home and abroad is focused on preeclampsia - eclampsia etiology and pathogenesis. overseas scholars think that preeclampsia (PE Preeclampsia) development can be summarized into two stages: the formation of the placenta and placental oxidative stress in non-performing . The process of oxidative stress in the placenta can release a range of placental factor, which causing endothelial cell damage and inflammatory response system, eventually led to a series of clinical preeclampsia symptoms and signs. The preeclampsia - eclampsia basic pathology is Vascular endothelial cell damage and inflammatory response system caused systemic small artery spasm . Some scholars even believe that pregnancy induced hypertension itself is the result of excessive maternal inflammatory response. PAR-2 is a member of the PARs family, since it may be involved in the development of many diseases, in recent years, more and more people pay attention on it. PAR-2 is the first time in1994, found in the mouse DNA sequence of a G protein-coupled receptorgene sequence,as protease activated receptors. Research has shown that PAR-2 is widespread in the cardiovascular tissues and organs rich in blood vessels. The amino terminal of the PAR-2 can be hydrolysis by serine proteases, thrombin and trypsin ,the new terminal can bind and activate receptor itself , Start the intracellular signal transduction to take participate in various physiological and pathophysiological processes of the tissues and organs. including inflammation, oxidative stress,angiogenesis,pain ,repair and so on.Therefore,the study of the expression of PAR-2 in placenta between in normal pregnancy and preeclampsia will help to understand and clarify the function of this receptor in the body's pathophysiological response .Meanwhile it will provides a new way of thinking on diagnosis and treatment.This study discusses the function and clinical significance of PAR-2 in preeclampsia pathogenesis through detecting the expression condition of PAR-2 in placentas of normal pregnant women and preeclampsia patients.The experiment collects preeclampsia patients(30 mild patients and 30 severepatients)and normal pregnant women(30 cases)from the Second Hospital of Jilin University,and adopts immunohistochemistry to detect the expression condition of PAR-2 in placenta tissues.Experimental data apply statistical software SPSS13.0 for statistical analysis.Among the groups compared with single analysis of variance (One-wayANOVA) ,and correlation analysis adopts Pearson correlation test.Experimental results show that: Both cell membrane and cytoplasmic of the placental trophoblast cells have the expression of brown-yellow staining granules, corresponding nucleus are blue. the average gray value of PAR-2 in the normal pregnant group are: 172.85±2.66, The average gray value of PAR-2 in the mild preeclampsia group are: 148.54±2.51,and the average gray value of PAR-2 in the severe preeclampsia group are: 131.14±2.95.The gray value expression of PAR-2 in preeclampsia group is significantly lower than that in the normal,that it to say ,the expression of PAR-2 in preeclampsia group is significantly higher than that in the normal(The gray value greater the positive weaker ;the gray value smaller the positive stronger). The difference is statistically significant (P<0.05); the expression in severe preeclampsia group is significantly higher than that in the mild preeclampsia group,and the difference is statistically significant(P<0.05).The expression of PAR-2 in placental tissues of normal pregnant women, mild preeclampsia patients and severe preeclampsia patients is gradually increased.In summary, the following conclusions can be obtained:PAR-2 in placental tissue of normal pregnant women show low or negative expression, which may be related to PAR-2 in the bodies is widely exists.The PAR-2 show high level expression in placenta tissues of preeclampsia patients and may be related with the pathogenesis of preeclampsia;the expression in placenta tissues of severe preeclampsia patients is significantly higher than that in patients with mild preeclampsia. indicating that it may have negative correlation with the severity. In-depth understanding of PAR-2 expression and significance in the placenta is essential value to the etiology and mechanism of the hypertensive disorders in pregnancy , at the same time, it will provides a new way of thinking for diagnosis and treatment of preeclampsia.
