This dissertation included three parts:Part one:Literature reviewA review about growing environment,phytochemical investigation and pharma-cological activities of plants belong to three genus:Blechnum, Woodwardia, Brainea was presented.Part two:Experimentation1. Chemical investigation of the rhizoma of Brainea insignis.With the isolation of the acetidin extract of the rhizoma of Brainea insignis by silica gel, we got eight compounds.By the technique of spectrum('H-NMR,13C-NMR,2D-NMR) and the method of analysis(TLC), they were confirmed as protocatechuic acid,4-(3,4-dihydroxyphenyl)-but-3-en-2-one, p-hydroxybenzoic acid, matteucinol-7-O-Glucopyranside, 5-O-caffeoyl shikimic acid, amentoflavone, Ponasterone A and Ponasteroside A. Besides,the structure of two monomers is not confirmed.2. The quantitative determination of protocatechuic acid and p-hydroxybenzoic acid by HPLC from Brainea insignis collected from different areaes.We established the quantitative method about protocatechuic acid and p-hydroxybenzoic acid by HPLC,and evaluate the content of these two componds from Brainea insignis collected from different areaes. Regression equations of protocatechuic acid and p-hydroxybenzoic acid were respectively:Y=4143.2X-3.275(r=0.9999), Y=5353.2X-0.6667(r=0.9999), which showed that the calibration curve was linear within the range of 0.0202μg-0.1414μg and 0.006μg-0.042μg for protocatechuic acid and p-hydroxybenzoic acid respectively. The content of protocatechuic acid ranges from 0.0752mg/g-0.1314mg/g and the content of p-hydroxybenzoic acid ranges from 0.0177mg/g-0.0535mg/g. The rusults showed that the methods were steady and reliable and can be used as the determination methods. 3. The quantitative determination of ponasteroside A and ponasterone A by HPLC from Brainea insignis. collected from different areaes.Based on the quantitative method established by our team, we determine the content of these two componds from Brainea insignis collected from different areaes. The content of ponasteroside A ranges from 2.4826 mg/g~4.4729 mg/g and the content of ponasterone A ranges from 0.1057 mg/g~0.2642 mg/g.4.The quantitative determination of total flavonoids in Brainea insignis. collected from different areaes.The HCl-Mg colorimetry was established to determine total flavonoids of Brainea insignis. And rutin was selected as reference substance. Rutin had a liner relationship with the absorbance in the range of 0.0115mg/ml~0.1261mg/ml and regression equation was as follow: Y=6.5952X-0.0299(r=0.9997). The content of total flavonoids ranged from 10.91%~12.34%. The rusults showed that the method was steady and reliable and can be used as the determination method.Part three:Summary and discussionThe chemical investigation and quantitative study on Brainea insignis provided evidence for further study and the relativity of the structure and activities.It also provided reference for reasonable clinical use of Chinese medicine Guanzhong.
