This paper studied on chemical composition, different parts of the total alkaloid content, antimicrobial acticity and anti-tumor activity in vivo and in vitro and other aspects of leaves of Clivia miniata Regel.On chemistry,three compounds were separated from the petroleum ether extraction and the total of alkaloids by using silica gel column chromatography and thin layer chromatography from the leaves of Clivia miniata Regel.and identified by 13C-NMR,1H-NMR,DEPT,ESI and the three compounds wereβ-sitosterol,Ly-corine,Clivonidine.Theβ-sitosterol was the first time isolated from the leaves of Clivia miniata Regel.The petroleum ether extraction was analyzed by GC-MS and 30 compounds were identified.Among them,the higher content of the composition was (Z,Z)-9,12-Octadecadienoic acid (linoleic acid) and the total content of 11538%,Hexanal (hexanal) was about the total content of 11,479%,Hexanoic acid (hexanoic acid) was the the total content of 8.123%,while the 9,12-Octadecadienoic acid, ethyl ester (ethyl linoleate) was the total content of 5.501%,n-Hexadecanoic acid (palmitic acid) was the content of 4.938%.Octanoic acid (tallow acid) was the content of 3.71%.when the leaves of Clivia miniata Regel were baked,it would send out a special taste,relating with volatile components.Different parts of the total content of alkaloids of Clivia miniata Regel were determined By UV spcctrophotometry.The results showed that The average content of the total alkaloids of different parts of Clivia miniata Regel were:Rhizome>Leaf>Root.On antimicrobial activity, the bacteria growth inhibition assay of experiments using Prayer-book method for ethanol extract of the leaves of Clivia miniata Regel and the antibacterial effect of the total alkaloids. The results showed that ethanol extract of the total alkaloids showed different inhibitory effect for E. coli ATCC8099 and.S. aureus ATCC6538, but not very obvious. The inhibitory effect of the total alkaloids on the E. coli ATCC8099 and S.aureus ATCC6538 were immeasurable effect relationship.In the 50mg/ml, the highest inhibition rate were 28.06% and 21.49%, respectively.The inhibition rate of ethanol extract on E. coli ATCC8099 and S. aureus ATCC6538 were 11.31% and 30.22%.On anti-tumor activity in vitro,the total of alkaloids of leaves of Clivia miniata Regel had a strong inhibitory effect on the 4 kinds of cultured tumor cells. They were NCI-H446,SGC-7901, MCF-7 and SMMC-7721 and showed a good dose-effect relationship.The IC50 values roled 48h for the 4 kinds of tumor cells were 38.55μg·L-1,12.08μg·mL-1,8.85μg·mL-1,7.04μg·mL-1 of the total of alkaloids of the leaves of Clivia miniata Regel.Compound 3 effected on SMMC-7721 cell proliferation without significant inhibition.On anti-tumor activity in vitro, the inhibitory effects of the total of alkaloids of Clivia miniata Regel on the growth of H22 tumor-bearing mice were detected. The results showed that the total of alkaloids of Clivia miniata Regel has antitumor effectivity. In the 34mg·kg-1, its inhibitory rate was 35.2%, compared with the negative control group, significant difference (P<0.05). While the total of alkaloids of Clivia miniata Regel on the immune organ has a certain impact.Dose at 136 mg·kg-1, the spleen index was significantly lower than the negative control group, showed significant differences (P<0.01).At the dose of 136mg·kg-1 and 68mg·kg-1. the thymus index was significantly lower compared with the negative control group, showed significant differences (P<0.01), compared with the positive control group also showed significant differences (P<0.01). White blood cell count compared with the negative control group and positive control group was not significant (P> 0.05).
