Chinese medicine coptidis rhizoma is the dry rhizoma of Coptis chinensis Franch., Coptis deltoidea C.Y. Cheng et Hsiao or Coptis teeta Wall., which are ranunculaceae plants. Historically, Emei area is the genuine producing area of coptidis rhizoma and coptdis deltoideae. Besides, there are C.omeiensis (Chen) C.Y. Cheng, and C.linearisepala T.Z. Wang et C.K. Hsieh. growing in Emei area. The subject researched the genuineness of Coptis chinensis in Emei area systematically via ecological factors, environmental factors and humanistic factors.1,According to the herbal textual research, we found that Sichuan is the genuine producing area of Coptis chinensis from ancient times, and Emei area is it's core areas.Before Ming Dynasty, there was artificial cultivation of Coptis chinensis in Emei area.2,The thesis summarized the long-term cultivation experiences of Coptis chinensis production base on Emei area including it's agricultural ecological environment, germplasm resources and cultivation techniques,and finally formulated the standard operation procedure of Coptis chinensis in Emei area under the guidance of GAP principles promulgated by SFDA.3,The pharmacognostic studies including microscopic and TLC identification. The results show that all of test materials are the dry rhizoma of Coptis chinensis Franch..Epiberberine, coptisine, palmatine and berberine were determined by HPLC methods. The results show that the berberine content of coptidis rhizoma in genuine producing area are more than non-genuine producing area. The berberine content of sample in Longdong, Xianfeng, Fuyou and Qili is more than Shizhu and Lichuan. The quality of coptidis rhizoma in Qili village in Emei is the best.4,HPLC fingerprint was established successfully and 10 samples are tested. The common mode for HPLC fingerprint of Coptis chinensis was established. The results of similarity evaluation between each test samples are 0.98~1.0, it shows that the quality of Coptis chinensis in Emei area is stable.5,The genomic DNA of Coptis chinensis collected from different areas were analyzed by RAPD-PCR methods.The PPB, Nei's genetic identity and genetic distance have been carried out and a UPGMA tree plot has been constructed. The results indicate that the species in Coptis chinensis have a closer genetic relationship. The genetic diversities of different geographic populations are not far different from that in single popolation. As a genuine medicinal materials, Coptis chinensis in Emei area has a stable genetic character.
