Dipsacus asper Wall., the Dipsacaceae plant, Perennial herb. Its dried root is a traditional Chinese herbal medicines. The Dipsacus asper Wall. Located in Wufeng County of Yichang city and Hefeng County in Enshi city was named because of its high quality, that is one of Enshi genuine regional drugs. In this study, the Dipsacus asper Wall. within the EnShi City, Hubei Province eight origine (Longping,Yuanbao,Yutangba embankments,The Se mine enriched area of Yutangba,Lvcongpo,Shuanghe,Taiping,Wuliping)for the material, from the morphological characteristics, photosynthetic characteristics, proxidase isozymes and DNA level of its genetic diversity,which is the basis of Dipsacus asper Wall.for the Identification and protection of the quality. The conclusions are as follows:1. Morphological characteristicsThe results showed that in root diameter, root length, lateral root number, stem length, stem edge number, stem diameter, leaf width ratio, and seed weight characteristics of the eight traits, the root diameter, root length, lateral root number and seed weight reached to a significant difference(P<0.05), accounting for half of all traits, including seed weight reached to a significant difference (P<0.01) levels. Integrated quality traits and cluster analysis of quantitative traits, the county Longping, Lichuang's Dipsacus asper Wall. element is divided into one group; Yutangba embankments, The Se mine enriched area of Yutangba in Enshi City, Shuanghe, Lvcongpo's Dipsacus asper Wall. for a class; Wuliping ,Taiping of Hefeng and the continued off into a class.2. Photosynthetic characteristicsUsing the ultraviolet absorption spectrophotometry measure the chlorophyll content of fresh leaves, the Se mine enriched area of Yutangba's chlorophyll a, chlorophyll b was the highest, followed by Hefeng Wuliping and Taiping.The material's net photosynthetic rate, dark respiration rate showed a diurnal variation of single peak curve, photosynthetic products accumulate mainly in the morning. The origin light saturation point between 291.67 689.71μmol/m2s; light compensation point in the 20.88 72.52μmol/m2s between the Hefeng Taiping's light saturation point of the highest, reaching 689.71μmol/m2s; Hefeng Wuliping's lowest light compensation point for 20.88μmol/m2s.The light saturation point and light compensation points of eight material are low, indicating that its suitable cultivation is half of the female half of the sun environment.Based on the above data,we can find that the authentic origin of Dipsacus asper Wall. in Hefeng Wuliping, Taiping and the Se mine enriched area of Yutangba are strongly in the chlorophyll content and photosynthetic capacity to the other five results. 3. Proxidase isozymesIn isozyme studies, Separation gel concentrations of the factors influencing the effect of band. In this study,using 9.0% separating gel concentration of peroxidase isozymes have the best bands. The peroxidase isozyme of Dipsacus asper Wall. total of 5 bands, Rf values between 0.35 to 0.55. Longping and Yutangba embankments of the band to the highest of 4 and Lvcongpo have one band only.According to differences in zymogram, the materials would be divided into 8 parts for two major categories. The first category include the Longping, Yutangba embankments, The Se mine enriched area of Yutangba, Hefeng Taiping; the second categories, including Lichuang, Lvcongpo , Shuanghe and Wuliping .4.Molecular markersThe best RAPD reaction system conditions for Dipsacus asper Wall. are:in 20μL reaction system to 50ng/μl template DNA concentration,1.0μmol/L concentration of primers, 2.5mmol/L of Mg2+concentration Annealing temperature of 39℃for the best. Reaction program was 94℃5minâ†'(94℃30sâ†'39℃40sâ†'72℃2min) 43cyclesâ†'72℃10minâ†'4℃maintained.With the best conditions, selected five polymorphic primers from fifty random primers.With the cluster Analysis, the materials will be divided into four parts.Groupâ… including Longping, Yuanbao; Groupâ…¡, including Yutangba embankments, The Se mine enriched area of Yutangba, Lvcongpo; Groupâ…¢including Taiping and Wuliping; Shuanghe of Enshi City is divided into a class of independent.5.EvaluationDiscussed the genetic diversity of Dipsacus asper Wall. in Enshi.The material of eight region has some difference in morphological traits such as leaf shape, light saturation point and other photosynthetic parameters, peroxidase isozyme characteristics and molecular level. Comprehensive classification results obtained are consistent, indicating that the combination of these evaluation methods is feasible.Based on the findings, eight materials can be divided into two group or five group,two group:the first category include the Longping, Yutangba embankments, The Se mine enriched area of Yutangba, Hefeng Taiping; the second categories, including Lichuang, Lvcongpo , Shuanghe and Wuliping.Five group: Longping and Yuanbao; Yutangba embankments and the Se mine enriched area of Yutangba; Hefeng Taiping and Wuliping; Lvcongpo and Shuanghe each for a class.According to authentic assessment of experimental results,look from the cluster analysis:the closer location of materials, the closer the genetic relationship, the smaller the differences between them.The root diameter, root length, lateral root number and seed weight of these traits teasel and the apparent of peroxidase isozyme between material are significant, it is recommended as the main parameters of Dipsacus asper Wall. in authentic assessment.The material of Taiping and Wuliping's photosynthetic capacity was stronger than other origin, consistent with the name of genuine regional drugs. Because the quality of medicines related to the environment and the special place of origin, so different habitats is the main reason for Dipsacus asper Wall. of different traits. In addition,the Se mine enriched area of Yutangba with higher chlorophyll a, b and its photosynthetic capacity just lowerthan the Taiping and Wuliping. It may be selenium can promote the elements of the chlorophyll synthesis. In the other study,selenium content of plant whether as a parameter yet further thought.
