Objective: Coronary Atherosclerotic Heart Disease (CHD) has become one of the most serious diseases that threatens human being's health. With the increasing of the living standard, the aging of population and the changing of diets and life-styles, CHD presents a trend of escalation. In our country, nearly twenty million people die of heart disease each year, accounting for one third of the total number of deaths. Therefore, the prevention and treatment of CHD, for example improving of lifestyle, eating low fat diet, losing weight, arouse extensive concern. The patients with hyperlipemia can have lower lipid treatment. At present, Western medicine therapy on patients with CHD has been widely used in clinical application. While traditional Chinese medicine therapy attracts more and more attention for the reason that the side-effect of western medicine is unnegligible in long therapeutic procedure. We have created the rat coronary atherosclerotic models and observed the effects of different dose of ShenShao oral liquid on these models. We have also observed different expressions of adiponectin and leptin by immunohistochemistry before and after the use of ShenShao oral liquid to research its mechanism and provide important basis for its effect on coronary atherosclerosis.Method:1 Feed 50 SD male rats adaptability for one week, and divide them into five groups randomly. Each group has ten rats. Groups of experimental animals are as follows: normal contorl group, atherosclerosis model group, low dose of shenshao oral liquid group, high dose of shenshao oral liquid group, simvastatin control group.2 Before modeling, rats in atherosclerosis model group, low dose of shenshao oral liquid group, high dose of shenshao oral liquid group and simvastatin control group were given intraperitoneal injection of VD3 (600,000 IU/kg/d) in three continuous days,and then fed with a high-fat diet for 8 weeks. Rats in normal contorl group were given intraperitoneal injection of the same dose of normal saline, then fed with normal diet for 8 weeks.In the ninth week, rats in low dose of shenshao oral liquid group were given Shenshao Oral Liquid 4ml/d orally, rats in high dose of shenshao oral liquid group were given Shenshao Oral Liquid 8ml/d orally, rats in simvastatin control group were given simvastatin 5mg/kg/d gavage, while rats in normal contorl group and atherosclerosis model group were given saline 4ml/d orally. Medicine time is from 9:00 to 10:00 in the morning, the period is 4 weeks. The total period of experiment is 12 weeks.3 Rats in each group are executed at the end of the twelfth week, then take intact aortas of the rats and stain them with HE and immunohistochemistry.4 The formation of atherosclerotic plaque of aorta is observed by HE. The expressions of adiponectin and leptin on endothelial cells of aorta are observed by immunohistochemistry.5 All the statistics are analyzed by SPSS 13.0.Results:1 HE shows that aorta of rats in atherosclerosis model group has unclear structure, coarse endangium and large area of incrassation and apophysis. The endothelial cells lacks and then lead to the formation of the plaque. We can see foam cells in the plaque, and amorphous substances, which include cholesterol crystal , necrotic tissues and a little fibrosis at the bottom of the plaque. We have observed migrated smooth muscle cells at the bottom of endangium, the endochylema of them stained light which seems to have lipids in them. The hyperplastic smooth muscle cells in tunica elastica ranged irregularly and migrated towards the endangium, and then formed lipide stripe.2 The expression of adiponectin on aortas of rats in each group: immunohistochemistry shows that adiponectin expresses at the endochylema of endothelial cells. The normal contorl group express some flagrant tan particles, staining scores 8.40±0.97. Compared with normal contorl group, the expressment of adiponectin in atherosclerosis model group decreases obviously, staining scores 3.00±0.82. The differences have statistical significance(p<0.05). Compared with atherosclerosis model group, the expressment of adiponectin in low dose of shenshao oral liquid group increases, staining scores 4.40±1.17. The differences have statistical significance(p<0.05). Compared with atherosclerosis model group and low dose of shenshao oral liquid group , the expressment of adiponectin in high dose of shenshao oral liquid group increases, staining scores 7.40±1.26. The differences have statistical significance(p<0.05). Compared with atherosclerosis model group, the expressment of adiponectin in simvastatin control group increases obviously, staining scores 7.30±1.42. The differences have statistical significance(p<0.05). Compared with high dose of shenshao oral liquid group, the differences have no statistical significance(p>0.05). 3 The expression of leptin on aortas of rats in each group: Leptin expresses at the endochylema of endothelial cells. The normal contorl group express a little flagrant tan particles, staining scores 2.70±0.67. Compared with normal contorl group, the expressment of leptin in atherosclerosis model group increases obviously, staining scores 8.50±0.53.The differences have statistical significance(p<0.05). Compared with atherosclerosis model group, the expressment of leptin in low dose of shenshao oral liquid group decreases, staining scores 6.80±1.03. The differences have statistical significance(p<0.05). Compared with atherosclerosis model group and low dose of shenshao oral liquid group, the expressment of leptin in high dose of shenshao oral liquid group decreases obviously, staining scores 4.00±0.82. The differences have statistical significance(p<0.05). Compared with atherosclerosis model group, the expressment of leptin in simvastatin control group decreases obviously, staining scores 4.10±1.14. The differences have statistical significance(p<0.05). Compared with high dose of shenshao oral liquid group, the differences have no statistical significance(p>0.05).Conclusion:1 The research shows that, the expression of adiponectin on aortas of rats in rat coronary atherosclerotic models decreases significantly, while the expression of leptin increases significantly.2 ShenShao oral liquid play a important role in coronary atherosclerosis by increasing the expression of adiponectin and decreasing the expression of leptin.3 This research shows that high dose of ShenShao oral liquid on rat coronary atherosclerosis is much more effective than high dose of ShenShao oral liquid.
